Herein, we realized the simultaneous online detection of six tobacco alkaloids (TM) by in-tube solid-phase microextraction (In-tube SPME) coupled with mass spectrometry by a rapid, sensitive, and matrix effect-free method requiring no chromatographic separation and only minimal sample pre-treatment. A poly (N-isopropylacrylamide-co-divinylbenzene-co-N, N’-methylenediacrylamide) [Poly (NIPAAm-co-DVB-co-MBAA)] monolithic column was designed according to the chemical structures of selected TM and used as an extraction medium engaging in hydrophobic, pi-pi, and hydrogen bonding interactions with analytes, allowing them to be effectively extracted. A number of important parameters were systematically optimized to achieve maximal extraction efficiency. The ion intensity of the TM signals obtained by in-tube SPME-MS were higher than the direct MS mode by about 400 folds with the signal-to-noise ratio improved by 2-7 folds. The detection limits of the six TM were determined as 1.99-4.06 ng g(-1), with good linearity with correlation coefficients exceeding 0.99 obtained under optimal extraction conditions. Besides, TA recoveries in cigarette tobacco spiked at three concentration levels were in the range of 76.4-100.2%, and the corresponding RSDs (n = 5) were obtained as 4.32-7.16%. The extraction performance of the poly (NIPAAm-co-DVB-co-MBAA) monolithic column was well reproducible, with intra- or inter-day precision RSDs determined not to exceed 7.38%. Finally, no marked matrix effects were observed when the developed method was applied to the analysis of both high-abundance and trace level TM in practical samples, and the above technique was therefore concluded to be well suited for the detection of TAs in cigarette tobacco or other products.

Background: During last recent years number of anti-tubulin agents were introduced for treatment of diverse kind of cancer. Despite of their potential in treatment of cancer, drug resistance and adverse toxicity such as peripheral neuropathy are some of the negative criteria of anti-tubulin agents. Methods: Twenty seven quinazoline derivatives were synthesized using a multicomponent reaction. The cytotoxicity of compounds 1-27 was tested in SRB assays employing five different human tumor cell lines. Effect of two of active compounds on tubulin polymerization was also checked using a commercially available assay kit. Molecular modelling studies were also performed using autodock tools software. Results: SRB assays showed that compounds 2, 9, 16 and 26, being highly cytotoxic with IC50 values ranging between 2.1 and 14.3 mu M. The possible mode of action of compounds, 2, 9, 16 and 26, and the taxol binding site of the protein tubulin, an important goal for antimitotic drugs, was also studied by molecular docking, which showed reasonable interactions with tubulin active site, followed by investigation of the effects of compounds 9 and 16 on the polymerization of tubulin. The results showed the tested compounds to be highly active as inducers of tubulin polymerization. Conclusion: Altogether, with respect to obtained results, it is attractive and beneficial to further investigation on quinazoline scaffold as antimitotic agents.

The photoswitching properties of three readily accesible benzoylpyridine hydrazones were investigated. Interestingly, replacing classical stirring with ultrasound wave activation results in pure thermodynamically less stable E isomer crystallization at room temperature. The studied benzoylpyridine hydrazones exhibit both P- and T-photochromic behaviour, depending on the benzoyl substituent, and improved addressability compared to most of the previously published pyridyl based hydrazones and 2-pyridylcarboxaldehyde acylhydrazones. Low activation entropy and calculated transition state geometry favour the inversion mechanism of their thermal isomerization rather than tautomerization followed by rotation recently found for pyridyl-hydrazone ester or nitrile rotary switches. The association behaviour of the nitro derivative during its thermal E-to-Z isomerization in highly polar DMSO indicates an important role of intermolecular hydrogen bonding in the thermal kinetics of benzoylpyridine-based hydrazone photoswitches. Moreover, the addition of triethylamine significantly accelerates the rate of Z-isomer thermal isomerization from days to seconds and could thus pave the way to fast pyridyl hydrazone T-type photochromic compounds in polar solvents. This study could therefore contribute to general knowledge related to the photochromic behaviour of hydrazones as an important class of modern photoswitches.

A series of halogenated imidazo[1,5-alpha]pyridines was prepared and their structural and optical properties investigated. The products were characterized by spectroscopic techniques and their optical properties discussed in relation to their chemical structure. We were able to triplicate the luminescence quantum yields (phi) in solution for three different imidazo[1,5-alpha]pyridine derivatives, depending on the halogen nature of the substituent on the imidazo[1,5-alpha]pyridine moiety. The effect of the halogen and of the geometrical and chemical characteristics of the molecular skeleton on the crystal packing was studied in the solid state by single crystal X-ray diffraction. At the same time, the possible presence of halogen bonds was evaluated through solid-state NMR analysis.

Background De novo lipogenesis (DNL) is the primary metabolic pathway synthesizing fatty acids from carbohydrates, protein, or alcohol. Our aim was to examine associations of in vivo levels of selected fatty acids (16:0, 16:1n7, 18:0, 18:1n9) in DNL with incidence of type 2 diabetes (T2D). Methods and findings Seventeen cohorts from 12 countries (7 from Europe, 7 from the United States, 1 from Australia, 1 from Taiwan; baseline years = 1970-1973 to 2006-2010) conducted harmonized individual-level analyses of associations of DNL-related fatty acids with incident T2D. In total, we evaluated 65,225 participants (mean ages = 52.3-75.5 years; % women = 20.4%62.3% in 12 cohorts recruiting both sexes) and 15,383 incident cases of T2D over the 9-year follow-up on average. Cohort-specific association of each of 16:0, 16:1n7, 18:0, and 18:1n9 with incident T2D was estimated, adjusted for demographic factors, socioeconomic characteristics, alcohol, smoking, physical activity, dyslipidemia, hypertension, menopausal status, and adiposity. Cohort-specific associations were meta-analyzed with an inverse-varianceweighted approach. Each of the 4 fatty acids positively related to incident T2D. Relative risks (RRs) per cohort-specific range between midpoints of the top and bottom quintiles of fatty acid concentrations were 1.53 (1.41-1.66; p< 0.001) for 16:0, 1.40 (1.33-1.48; p< 0.001) for 16:1n-7, 1.14 (1.05-1.22; p = 0.001) for 18:0, and 1.16 (1.07-1.25; p< 0.001) for 18:1n9. Heterogeneity was seen across cohorts (I-2 = 51.1%-73.1% for each fatty acid) but not explained by lipid fractions and global geographical regions. Further adjusted for triglycerides (and 16:0 when appropriate) to evaluate associations independent of overall DNL, the associations remained significant for 16:0, 16:1n7, and 18:0 but were attenuated for 18:1n9 (RR = 1.03, 95% confidence interval (CI) = 0.94-1.13). These findings had limitations in potential reverse causation and residual confounding by imprecisely measured or unmeasured factors. Conclusions Concentrations of fatty acids in the DNL were positively associated with T2D incidence. We herein report the fabrication of a bifunctional iron nanocomposite catalyst, in which two catalytically active sites of Fe-N-x and Fe phosphate, as oxidation and Lewis acid sites, were simultaneously integrated into a hierarchical N,P-dual doped porous carbon. As a bifunctional catalyst, it exhibited high efficiency for direct oxidative cleavage of alkenes into ketones or their oxidation into 1,2-diketones with a broad substrate scope and high functional group tolerance using TBHP as the oxidant in water under mild reaction conditions. Furthermore, it could be easily recovered for successive recycling without appreciable loss of activity. Mechanistic studies disclose that the direct oxidation of alkenes proceeds via the formation of an epoxide as intermediate followed by either acid-catalyzed Meinwald rearrangement to give ketones with one carbon shorter or nucleophilic ring-opening to generate 1,2-diketones in a cascade manner. This study not only opens up a fancy pathway in the rational design of Fe-N-C catalysts but also offers a simple and efficient method for accessing industrially important ketones and 1,2-diketones from alkenes in a cost-effective and environmentally benign fashion.

Triazole-based ligands, tris (triazolyl)methanol (Htbtm), bis (triazolyl)-phenylmethanol (Hbtm), and phenyl (pyridin-2-yl)(triazolyl)methanol (Hpytm), with differences in ligand denticity (i.e., bidentate and tridentate) and type of N donors (i.e., triazole and pyridine) were functionalized onto a silica support to produce the corresponding SiO2-L (L = tbtm, btm, pytm). Subsequent reactions with Pd (CH3COO)(2) in CH2Cl2 yielded Pd/SiO2-L. ICP-MS reveals that Pd loadings are higher with increased N loadings, resulting in the following trend: Pd/SiO2-tbtm (0.83 mmol Pd g(-1)) > Pd/SiO2-btm (0.65 mmol Pd g(-1)) ~ Pd/SiO2-pytm (0.63 mmol Pd g(-1)). Meanwhile, TEM images of the used Pd/SiO2-L catalysts after the first catalytic cycle show that the mean size of Pd NPs is highest with Pd/SiO2-pytm (8.5 +/- 1.5 nm), followed by Pd/SiO2-tbtm (6.4 +/- 1.6 nm) and Pd/SiO2-btm (4.8 +/- 1.3 nm). Based on TONs, catalytic studies toward aerobic oxidation of benzyl alcohol to benzaldehyde at 60 degrees C in EtOH showed that Pd/SiO2-pytm possessed the most active surface Pd(0) atoms, most likely as a result of more labile properties of the pyridine-triazole ligand compared to tris- and bis (triazolyl) analogs. ICP-MS and TEM analysis of Pd/SiO2-btm indicate minimal Pd leaching and similar average Pd NPs sizes after 1(st) and 5(th) catalytic runs, respectively, confirming that SiO2-btm is an efficient Pd NPs stabilizer. The Pd/SiO2-btm catalyst was also active toward aerobic oxidation of various benzyl alcohol derivatives in EtOH and could be reused for at least 7 reaction cycles without a significant activity loss.
