N-(Pyrid-2-yl)-substituted azolium and pyridinium salts, precursors for hybrid NHC-containing ligands, were obtained with excellent regioselectivity, employing a deoxygenative CH-functionalization of pyridine-N-oxides with substituted imidazoles, thiazoles, and pyridine. Unlike the traditional SNAr-based methods, this approach provides high yields for substrates bearing substituents of different electronic nature. The utility of azolium and pyridinium salts thus prepared was also highlighted by the synthesis of pyridyl-substituted imidazolyl-2-thione, benzodiazepine as well as 2-aminopyridines.

A practical heterogeneous palladium-catalyzed carbonylative Suzuki coupling of aryl iodides with arylboronic acids under carbon monoxide gas-free conditions has been developed using a bidentate phosphino-functionalized magnetic nanoparticle-immobilized palladium(II) complex as catalyst. Formic acid was utilized as the carbon monoxide source with dicyclohexylcarbodiimide as the activator, and a wide variety of biaryl ketones were generated in moderate to high yields. The new heterogeneous palladium catalyst can be prepared via a simple procedure and can easily be separated from a reaction mixture by simply applying an external magnet and recycled up to 10 times without any loss of activity.

The interaction between pyridoxal hydrochloride (HQ) and apoCopC was investigated using Fourier transform infrared spectroscopy (FTIR), isothermal titration calorimetry (ITC), circular dichroism (CD), fluorescence spectroscopy, three-dimensional (3D) fluorescence spectroscopy, fluorescence lifetime, TNS fluorescence and docking methods. FTIR, CD, TNS fluorescence and fluorescence lifetime experiments suggested that the apoCopC conformation was altered by HQ with an increase in the random coil content and a reduction in the beta-sheet content. In addition, the data from fluorescence spectroscopy, 3D fluorescence spectroscopy and molecular docking revealed that the binding site of HQ was located in the hydrophobic area of apoCopC, and a redshift of the HQ fluorescence spectra was observed. Furthermore, ITC and fluorescence quenching data manifested that the binding ratio of HQ and apoCopC was 1:1, and the forming constant was calculated to be (7.06 +/- 0.21) x 10(5) M-1. The thermodynamic parameters Delta H and Delta S suggested that the formation of a CopC-HQcomplex depended on the hydrophobic force. Furthermore, the average binding distance between tryptophan in apoCopC and HQ was determined by means of Forster non-radioactive resonance energy transfer and molecular docking. The results agreed well with each other. As a redox switch in the modulation of copper, the interaction of apoCopC with small molecules will affect the action of the redox switch. New Delhi metallo-beta-lactamase-1 (NDM-1) is capable of hydrolyzing nearly all beta-lactam antibiotics, posing an emerging threat to public health. There are currently less effective treatment options for treating NDM-1 positive superbug, and no promising NDM-1 inhibitors were used in clinical practice. In this study, structure-activity relationship based on thiosemicarbazone derivatives was systematically characterized and their potential activities combined with meropenem (MEM) were evaluated. Compounds 19bg and 19bh exhibited excellent activity against 10 NDM-positive isolate clinical isolates in reversing MEM resistance. Further studies demonstrated compounds 19bg and 19bh were uncompetitive NDM-1 inhibitors with Ki = 0.63 and 0.44 mu mol/L, respectively. Molecular docking speculated that compounds 19bg and 19bh were most likely to bind in the allosteric pocket which would affect the catalytic effect of NDM-1 on the substrate meropenem. Toxicity evaluation experiment showed that no hemolysis activities even at concentrations of 1000 mg/mL against red blood cells. In vivo experimental results showed combination of MEM and compound 19bh was markedly effective in treating infections caused by NDM-1 positive strain and prolonging the survival time of sepsis mice. Our finding showed that compound 19bh might be a promising lead in developing new inhibitor to treat NDM-1 producing superbug. A new fluorometric chemodosimeter 2-amino-3-(((E)-3-(1-phenylimidazo[1,5-]pyridin-3-yl)benzylidene)amino)maleonitrile (BPI-MAL) has been designed and synthesized for sensing hypochlorite. BPI-MAL showed a selective turn-on fluorescence for ClO- through hypochlorite-promoted de-diaminomaleonitrile reaction. It also could detect ClO- in the presence of various competitive anions including reactive oxygen species. Interestingly, sensor BPI-MAL was successfully applied as a fluorescent test kit for ClO- determination. The sensing property and mechanism of BPI-MAL toward ClO- were studied by fluorescence and UV-vis spectroscopy, NMR titration and DFT calculations.

A series of two new mono- and one binuclear m-nitrato bridged copper(II) complexes [Cu(L)(HL)]ClO4 (1), [Cu(HL)(NO3)(H2O)](2)NO3 center dot H2O (2) and [Cu-2(L-2)(mu-NO3)(2)] (3), with an unsymmetrical NNO donor Schiff base (HL) have been synthesized and characterized by elemental analysis, FTIR, CV, UV-vis and EPR spectroscopy. Their molecular structures were also determined by single crystal X-ray crystallography. In the binuclear complex 3, the Cu center dot center dot center dot Cu distance is 3.494 angstrom. In 1, 2 and 3, the Cu(II) centers have distorted square pyramidal geometry (tau(5) = 0.05-0.17). Evidence of weak pi center dot center dot center dot pi stacking intermolecular interactions along with other non-covalent interactions (hydrogen bonding) was observed by analyzing the respective crystal structures of the complexes. Thus, these hydrogen bonds, pi center dot center dot center dot pi stacking interactions and other weak intermolecular interactions establish in the form of supramolecular architectures a crystalline network environment. The non-covalent interactions were also investigated by employing Hirshfeld Analysis. The room temperature magnetic moments of the mononuclear complexes are less than the spin only values which are indicative of small interactions. Also, significant magnetic interactions were not exhibited by binuclear copper(II) complex 3 in the variable temperature magnetic measurements. The X-band EPR spectra of all three complexes exhibit copper(II) hyperfine structures as well as zero-field splitting which are appropriate for the triplet states of dimers. In complexes 1 and 2, the presence of pseudo dipolar interactions is proposed. Quantum chemical calculations (DFT) were carried out on complexes 1-3 to explore the electronic and spectral properties of these newly synthesized complexes. These complexes show significant antiproliferative and SOD activity. The SOD activity measured in terms of kMcCF is in the range 4.94-12.31 (mol L)(-1)s(-1))x 10(4). (C) 2021 Elsevier Ltd.
