A highly diastereoselective, visible-light-induced [3 + 2] cycloaddition between N-sulfonyl cyclopropylamines I (R = Ph, 4-MeC6H4, 2-naphthyl, 3-pyridinyl, etc.) and electron-deficient olefins R1R2C:CHR3 (R1 = EtO2C, CN, PhCO, PhNHCO, etc., R2 = H, Me, R3 = H; R1 = R2 = EtO2C, R3 = Me; R1 = Ph, R2 = H, R3 = MeO2C; etc.) is reported. The reactions proceed via the oxidation of a sulfonamide aza-anion by an organic photocatalyst to generate a nitrogen-centered radical. Strain-induced ring opening and intermol. addition to the olefin generate an intermediate carbon-centered radical that is reduced to an anion prior to 5-exo cyclization. This enables a highly diastereoselective construction of trans-cyclopentanes II possessing synthetically useful functional groups.

Indoleamine-2,3-dioxygenase-1 (IDO1) has emerged as a target of significant interest to the field of cancer immunotherapy, as the upregulation of IDO1 in certain cancers has been linked to host immune evasion and poor prognosis for patients. In particular, IDO1 inhibition is of interest as a combination therapy with immune checkpoint inhibition. Through an Automated Ligand Identification System (ALIS) screen, a diamide class of compounds was identified as a promising lead for the inhibition of IDO1. While hit 1() possessed attractive cell-based potency, it suffered from a significant right-shift in a whole blood assay, poor solubility, and poor pharmacokinetic properties. Through a physicochem. property-based approach, including a focus on lowering AlogP98 via the strategic introduction of polar substitution, compound 13 was identified bearing a pyridyl oxetane core. Compound 13() demonstrated improved whole blood potency and solubility, and an improved pharmacokinetic profile resulting in a low predicted human dose.

FAH domain containing protein 1 (FAHD1) acts as oxaloacetate decarboxylase in mitochondria, contributing to the regulation of the tricarboxylic acid cycle. Guided by a high-resolution X-ray structure of FAHD1 liganded by oxalate, the enzymic mechanism of substrate processing is analyzed in detail. Taking the chem. features of the FAHD1 substrate oxaloacetate into account, the potential inhibitor structures are deduced. The synthesis of drug-like scaffolds afforded first-generation FAHD1-inhibitors with activities in the low micromolar IC50 range. The investigations disclosed structures competing with the substrate for binding to the metal cofactor, as well as scaffolds, which may have a novel binding mode to FAHD1.

Increasing the remoteness of the donor N-atom from the bridgehead results in large differences in the coordination of the Li+ cations by the (8-quinolyl)aluminate anions compared to 2-quinolyl or 2-pyridyl counterparts. The results are of potential interest in understanding how the coordination sites of ligands of this type can be tuned for the coordination requirements of specific metal centers.

An efficient approach for divergent synthesis of primary, secondary, and tertiary amines, e.g., I via the merger of controllable cleavage of triazines and site-selective functionalization was disclosed. A number of imidazo [1,2-a]pyridines-containing primary and secondary aromatic amines, and trialkylamines were facilely accessed with a wide range of functional groups. Importantly, the obtained aromatic amines could be incorporated with important bioactive units and converted into many valuable nitrogen-containing mols. In addition, the results revealed that Hofmann-Martius type rearrangement exhibits excellent o- and p-selectivity and goes through an intermol. process.

Four novel compounds (S-DCM-1O, S-DCM-2O, S-DCM-3O, and S-DCM-4O) based on sulfur-substituted dicyanomethylene-4H-chromene (S-DCM) and diarylethene were synthesized. The detailed investigations on the fluorescence spectra, absorption spectra, and time-dependent d. functional theory calculations (DFT/TD-DFT) were presented. The encapsulation of mPEG2000-DSPE effectively increased the solubility of all the as-prepared samples. The results of TEM showed that the nanoparticles had a good dispersion coefficient All the nanomaterials (S-DCM-1O@PEG, S-DCM-2O@PEG, S-DCM-3O@PEG, and S-DCM-4O@PEG) exhibited near-IR emission (650-900 nm) and thus reduced light damage and light scattering for cell imaging in biol. applications. Furthermore, PLL (poly-L-lysine hydrobromide) was used to modify the surface of nanoparticles to facilitate easy uptake by A549 cells by changing their electronegativity. The results suggested that S-DCM-2O@PEG@PLL was a highly suitable candidate for cell imaging because of its low toxicity and excellent biocompatibility.

Wang, Yishou;Li, Shichen;Wang, Xinfeng;Yao, Yiming;Feng, Lei;Ma, Chen research published 《 A multi pathway coupled domino strategy: I2/TBHP-promoted synthesis of imidazopyridines and thiazoles via sp3, sp2 and sp C-H functionalization》, the research content is summarized as follows. I2/TBHP-promoted, one-pot, multi pathway synthesis of imidazopyridines and thiazoles was achieved through readily available ethylarenes, ethylenearenes and ethynearenes. I2/TBHP as an initiator and oxidant was used to realize the C-H functionalization of this domino reaction. Simple and available starting materials, wide range of functional group tolerance, high potential for drug activity of the products and application in production were the advantageous features of this method.

Wang, Yi-Dong;Liu, Jin-Kui;Yang, Xue-Jie;Gong, Jun-Fang;Song, Mao-Ping research published 《 Chiral (Pyridine)-(Imidazoline) NCN’ Pincer Palladium(II) Complexes: Convenient Synthesis via C-H Activation and Characterization》, the research content is summarized as follows. A series of aryl-based chiral NCN’ pincer palladium(II) complexes 4ak were synthesized via C-H activation of the (pyridyl)-(imidazolinyl)benzene ligands, which proceeded smoothly with PdCl2 as the palladium source in the presence of Et3N or NaHCO3 base in toluene under reflux. All of the complexes have been characterized by elemental anal. and 1H and 13C{1H} NMR spectroscopy. In addition, the mol. structures of complexes 4a, 4c, and 4f have been determined by x-ray single-crystal diffraction anal. The obtained chiral Pd pincers were applied to catalytic asym. reactions of acrylonitrile and dichloroacetonitrile with sulfonimines. With a catalyst loading of 5 mol % and the assistance of AgOAc, these complexes acted as active but moderately stereoselective catalysts for the aforementioned reactions.
