A three-descriptor quantitative structure-activity/toxicity relationship (QSAR/QSTR) model was developed for the skin permeability of a sufficiently large data set consisting of 274 compounds, by applying support vector machine (SVM) together with genetic algorithm. The optimal SVM model possesses the coefficient of determination R-2 of 0.946 and root mean square (rms) error of 0.253 for the training set of 139 compounds; and a R-2 of 0.872 and rms of 0.302 for the test set of 135 compounds. Compared with other models reported in the literature, our SVM model shows better statistical performance in a model that deals with more samples in the test set. Therefore, applying a SVM algorithm to develop a nonlinear QSAR model for skin permeability was achieved.

BACKGROUND There is a close relationship between certain ‘aging markers’ in wine and the wine’s age. This study aimed to characterize all extracted aging markers in Chinese rice wine and distinguish the ages of Chinese rice wine using principal component analysis (PCA) and cluster analysis (CA). RESULTS A total of 49 potential aging markers (P <= 0.05*) were extracted from 71 volatile organic compounds (VOCs) in Chinese rice wine across a range of seven different ages. Of all 49 extracted aging markers, all furans, 2/3 aldehydes and ketones maintained significantly increasing levels with age (P <= 0.01**), especially sotolon (0.981**) and acetophenone (0.951**). On the other hand, all sulfides decreased significantly (P <= 0.01**). Changes in vanillin, guaiacol, 4-vinylguaiacol (4-VG) and 4-ethylguaiacol (4-EG) also suggested a potential synthesis during the aging process. The results of PCA and CA demonstrated that Chinese rice wines with different ages could be clearly distinguished from each other, which was consistent with the evolution of the 49 aging markers during the aging process. CONCLUSION These 49 potential 'aging markers' successfully distinguished ages using PCA and CA. Our results therefore throw light on the characterization of VOCs during Chinese rice wine aging, and provide a quantitative basis for discriminating the ages of Chinese rice wine. A pyrimidine moiety exhibiting a wide range of pharmacological activities has been employed in the design of privileged structures in medicinal chemistry. To prepare libraries of novel heterocyclic compounds with potential biological activities, a series of novel 2-(pyridin-2-yl) pyrimidine derivatives were designed, synthesized and their biological activities were evaluated against immortalized rat hepatic stellate cells (HSC-T6). Fourteen compounds were found to present better anti-fibrotic activities than Pirfenidone and Bipy55 ‘ DC. Among them, compounds ethyl 6-(5-(p-tolylcarbamoyl)pyrimidin-2-yl)nicotinate (12m) and ethyl 6-(5-((3,4-difluorophenyl)carbamoyl)pyrimidin-2-yl)nicotinate (12q) show the best activities with IC50 values of 45.69 mu M and 45.81 mu M, respectively. Furthermore, the study of anti-fibrosis activity was evaluated by Picro-Sirius red staining, hydroxyproline assay and ELISA detection of Collagen type I alpha 1 (COL1A1) protein expression. Our study showed that compounds 12m and 12q effectively inhibited the expression of collagen, and the content of hydroxyproline in cell culture medium in vitro, indicating that compounds 12m and 12q might be developed the novel anti-fibrotic drugs.

An efficient acid-catalyzed condensation between substituted benzohydrazides and 2,3,5,6-tetrafluoroterephthalic acid to form 1,4-bis(5-aryl-1,3,4-oxadiazol-2-yl)-2,3,5,6-tetrafluorobenzenes is reported. The products were isolated in 74-87% yield and were characterized by(1)H NMR, IR, and mass spectra.

We report a mild palladium-catalyzed selective dehydrogenation of 4-alkylpyridines that exploits their soft enolization to alkylidene dihydropyridines using allyl chloroformate under metal-free conditions. Treatment of these intermediates with a palladium catalyst liberates an alkylpyridylic anion and an allylpalladium(II) intermediate, which combine and undergo beta-hydride elimination to install a double bond. Importantly, the formation of gas byproducts makes the process very practical. Furthermore, the reaction tolerates a broad range of functional groups, including many that can be dehydrogenated using similar palladium-catalyzed reactions, and is selective for 4-alkylpyridines even in substrates bearing multiple pyridylic positions.

Three novel ruthenium nitrosyl complexes [Ru(NO)Cl-3(InicMe)(2)] (1b), [RuNOCl3(NicEt)(2)] (1c) and [RuNOCl3(NicMe)(2)] (1d) (InicMe = methyl isonicotinate, NicEt = ethyl nicotinate, NicMe = methyl nicotinate)were prepared and crystal structure of the complexes were determined by single crystal XRD analysis. In all complexes, the organic ligands are coordinated by a pyridine nitrogen atom and located in trans-position each to other and in cis-position to NO group. In the crystal package of all compounds stacking interactions of two types were determined: pi(arene)-pi(arene) and pi(COO)-pi(arene) stacking. Finally, cytotoxicity of the compounds was tested on Hep2 and HepG2 cell lines. In the set of similar compounds mer-[RuNO(L)(2)Cl-3] (L = Py, gamma-Pic, beta-Pic, Inic-Alk, Nic-Alk), complexes with iso-nicotinic acid esters are the most toxic, while nicotinic acid derivatives are less toxic and compared with pyridine complex. Herein, we realized the simultaneous online detection of six tobacco alkaloids (TM) by in-tube solid-phase microextraction (In-tube SPME) coupled with mass spectrometry by a rapid, sensitive, and matrix effect-free method requiring no chromatographic separation and only minimal sample pre-treatment. A poly (N-isopropylacrylamide-co-divinylbenzene-co-N, N’-methylenediacrylamide) [Poly (NIPAAm-co-DVB-co-MBAA)] monolithic column was designed according to the chemical structures of selected TM and used as an extraction medium engaging in hydrophobic, pi-pi, and hydrogen bonding interactions with analytes, allowing them to be effectively extracted. A number of important parameters were systematically optimized to achieve maximal extraction efficiency. The ion intensity of the TM signals obtained by in-tube SPME-MS were higher than the direct MS mode by about 400 folds with the signal-to-noise ratio improved by 2-7 folds. The detection limits of the six TM were determined as 1.99-4.06 ng g(-1), with good linearity with correlation coefficients exceeding 0.99 obtained under optimal extraction conditions. Besides, TA recoveries in cigarette tobacco spiked at three concentration levels were in the range of 76.4-100.2%, and the corresponding RSDs (n = 5) were obtained as 4.32-7.16%. The extraction performance of the poly (NIPAAm-co-DVB-co-MBAA) monolithic column was well reproducible, with intra- or inter-day precision RSDs determined not to exceed 7.38%. Finally, no marked matrix effects were observed when the developed method was applied to the analysis of both high-abundance and trace level TM in practical samples, and the above technique was therefore concluded to be well suited for the detection of TAs in cigarette tobacco or other products.
