New copper(II) hydrazone complexes with (Z)-2-(phenyl(2-(pyridin-2-yl)hydrazono)methyl)pyridine (L) were synthesized and characterized using various physicochemical methods. The geometries of the complexes can be classified as mononuclear and binuclear. The complex 1, [Cu(L)(Cl)(2)], is mononuclear whereas the solid-state structure of complex 2 contain a mixture of co-crystals of the mono- and binuclear complexes 2a, [Cu(L)(H2O)(SO4)], and 2b, [Cu-2(L)(2)(mu-SO4)(2)]. The unit cell of 2 contains two units of the mononuclear complex 2a and one units of the binuclear complex 2b. The copper atoms contained in all the mono- and binuclear complexes are in a distorted square pyramidal geometry. The present study indicates that complexes having different nuclearities and geometries can be achieved by changing the synthetic conditions and methods. Variable temperature magnetic susceptibility measurements of the complexes have shown the presence of weak anti-ferromagnetic interactions. These interactions are mediated by intermolecular hydrogen bonding in 1 and through a symmetric sulfate bridge in 2. The EPR spectra in the polycrystalline state for 1 and 2 exhibited a broad signal at similar to 2.149 due to spin-spin interactions between two copper(II) ions. The cyclic voltammograms of complexes 1 and 2 in DMSO gave two irreversible redox waves. Density functional theory (DFT) calculations were evaluated in the study, involving the molecular specification with the use of B3LYP/LANL2DZ formalism for the copper atoms and B3LYP/6-31G for the remaining atoms. Both complexes catalyzed the dismutation of superoxide (O-2(center dot-)). Furthermore, the copper complexes and the ligand were tested to explore their anticancer properties. Promising cytotoxicity of the synthesized compounds was observed against the selected cancerous cell lines of neuroblastoma, lung carcinoma, hepatocellular carcinoma and breast cancer. A series of two new mono- and one binuclear m-nitrato bridged copper(II) complexes [Cu(L)(HL)]ClO4 (1), [Cu(HL)(NO3)(H2O)](2)NO3 center dot H2O (2) and [Cu-2(L-2)(mu-NO3)(2)] (3), with an unsymmetrical NNO donor Schiff base (HL) have been synthesized and characterized by elemental analysis, FTIR, CV, UV-vis and EPR spectroscopy. Their molecular structures were also determined by single crystal X-ray crystallography. In the binuclear complex 3, the Cu center dot center dot center dot Cu distance is 3.494 angstrom. In 1, 2 and 3, the Cu(II) centers have distorted square pyramidal geometry (tau(5) = 0.05-0.17). Evidence of weak pi center dot center dot center dot pi stacking intermolecular interactions along with other non-covalent interactions (hydrogen bonding) was observed by analyzing the respective crystal structures of the complexes. Thus, these hydrogen bonds, pi center dot center dot center dot pi stacking interactions and other weak intermolecular interactions establish in the form of supramolecular architectures a crystalline network environment. The non-covalent interactions were also investigated by employing Hirshfeld Analysis. The room temperature magnetic moments of the mononuclear complexes are less than the spin only values which are indicative of small interactions. Also, significant magnetic interactions were not exhibited by binuclear copper(II) complex 3 in the variable temperature magnetic measurements. The X-band EPR spectra of all three complexes exhibit copper(II) hyperfine structures as well as zero-field splitting which are appropriate for the triplet states of dimers. In complexes 1 and 2, the presence of pseudo dipolar interactions is proposed. Quantum chemical calculations (DFT) were carried out on complexes 1-3 to explore the electronic and spectral properties of these newly synthesized complexes. These complexes show significant antiproliferative and SOD activity. The SOD activity measured in terms of kMcCF is in the range 4.94-12.31 (mol L)(-1)s(-1))x 10(4). (C) 2021 Elsevier Ltd. A new poly (vinyl chloride) (PVC) membrane electrode using 2-benzoylpyridine semicarbazone as membrane carrier with dioctylphthalate as plasticizer and sodium tetraphenylborate (NaTBP) as anion excluder has been fabricated and investigated as Zn(II)-selective electrode. Best potential response is observed for the composition PVC 30%, plasticizer 58%, NaTBP 8% and ionophore 4% (w/w). The sensor showed a linear stable response over a concentration range of 1.0 x 10(-2)-4.56 x 10(-6) M with a detection limit of 2.28 x 10(-6) M and a response time <10 s. Expanding the chemical diversity of metal complexes provides a robust platform to generate functional bioactive reagents. To access an excellent repository of metal-based compounds for probe/drug discovery, we capitalized on the rich chemistry of gold to create organometallic gold(III) compounds by ligand tuning. We obtained novel organogold(III) compounds bearing a 1,2-bis(diphenylphosphino)benzene ligand, providing structural diversity with optimal physiological stability. Biological evaluation of the lead compound AuPhos-89 demonstrates mitochondrial complex I-mediated alteration of the mitochondrial electron transport chain (ETC) to drive respiration and diminish cellular energy in the form of adenosine triphosphate (ATP). Mechanism-of-action efforts, RNA-Seq, quantitative proteomics, and NCI-60 screening reveal a highly potent anticancer agent that modulates mitochondrial ETC. AuPhos-89 inhibits the tumor growth of metastatic triple negative breast cancer and represents a new strategy to study the modulation of mitochondrial respiration for the treatment of aggressive cancer and other disease states where mitochondria play a pivotal role in the pathobiology.

P-glycoprotein (P-gp)-mediated multidrug resistance (MDR) is a major obstacle to successful chemotherapy for leukemia. In this study, a series of thiosemicarbazone-containing compounds (4a-b, 7a-q) were synthesized. Biological evaluation showed that the most active compound 7e displayed potent anti-leukemia activity against P-gp overexpressing drug-resistant K562/A02 cells, with an IC50 value of 0.44 mu M. Notably, compound 7e exhibited a selective killing effect on K562/A02 cells by dose-dependently increasing the intracellular levels of reactive oxygen species (ROS), thus exerting a potential collateral sensitivity (CS)-promoting effect in vitro. Moreover, compound 7e could inhibit HDAC1 and HDAC6, and induce the apoptosis of K562/A02 cells by increasing the expression of Bax, decreasing Bcl-2 protein level, and promoting the cleavage of caspase-3 and PARP, respectively. Overall, 7e may be a potential anti-cancer agent against drug-resistant myelogenous leukemia.

Objective: To overcome the disadvantages of cisplatin, numerous platinum (Pt) complexes have been prepared. However, the anticancer activity and mechanism of Pt(II) complexed with 2-benzoylpyridine [Pt(II)-Bpy]: [PtCl2(DMSO)L] (DMSO = dimethyl sulfoxide, L = 2-benzoylpyridine) in cancer cells remain unknown. Methods: Pt(II)-Bpy was synthesized and characterized by spectrum analysis. Its anticancer activity and underlying mechanisms were demonstrated at the cellular, molecular, and in vivo levels. Results: Pt(II)-Bpy inhibited tumor cell growth, especially HepG2 human liver cancer cells, with a half-maximal inhibitory concentration of 9.8 +/- 0.5 mu M, but with low toxicity in HL-7702 normal liver cells. Pt(H)Bpy induced DNA damage, which was demonstrated through a marked increase in the expression of cleaved-poly (ADP ribose) polymerase (PARP) and gamma-H2A histone family member X and a decrease in PARP expression. The interaction of Pt(II)-Bpy with DNA at the molecular level was most likely through an intercalation mechanism, which might be evidence of DNA damage. Pt(II)-Bpy initiated cell cycle arrest at the S phase in HepG2 cells. It also caused severe loss of the mitochondria( membrane potential; a decrease in the expression of caspase-9 and caspase-3; an increase in reactive oxygen species levels; the release of cytochrome c and apoptotic protease activation factor; and the activation of caspase-9 and caspase-3 in HepG2 cells, which in turn resulted in apoptosis. Meanwhile, changes in p53 and related proteins were observed including the upregulation of p53, the phosphorylation of p53, p21, B-cell lymphoma-2-associated X protein, and NOXA; and the down-regulation of B-cell lymphoma 2. Moreover, Pt(II)-Bpy displayed marked inhibitory effects on tumor growth in the HepG2 nude mouse model. Conclusion: Pt(II)-Bpy is a potential candidate for cancer chemotherapy.

Sulfur-containing species are essential in the composition and the metabolism of the organisms, thus developing a full set of implements to cover all of them is still a favorable choice. Herein, we chose imidazo [1,5-alpha] pyridine moiety as the basic fluorophore for the detection of sulfite, and preliminarily completed the toolset since biothiols (GSH, Cys, Hcy), H2S, and PhSH could be detected by sensors based on the same backbone. The designed sensor, IPD-SFT, with structural novelty and large Stokes shift (130 nm), indicated the most attractive advantages of remarkably rapid response period (within 1 min) and high selectivity for sulfite from all the sulfurcontaining species. Other practical properties included high sensitivity (LOD = 50 nM) and wide pH adaptability (5.0-11.0). Furthermore, IPD-SFT could monitor both exogenous and endogenous sulfite. It not only raised a potential tool for sulfite detection, but also preliminarily completed the toolset for all the sulfur-containing species. The development of such toolsets might reveal the sulfur-containing metabolism and corresponding physiology and pathological procedures.
