Three asymmetric tridentate acyl hydrazone Schiff base ligands namely L1, L2 and L3 were prepared via condensation of 4-methoxybenzohydrazide with picolinaldehyde, 1-(pyridin-2-yl)ethanone and phenyl(pyridin-2-yl)methanone respectively. Three bio-relevant mononuclear zinc(ii) complexes [Zn(L1)Cl-2]2H(2)O (1), [Zn(L2)Cl-2] (2) and [Zn(L3)Cl-2] (3) were synthesized by treatment of zinc(ii) chloride with the corresponding Schiff base ligands and characterised by the usual physicochemical techniques. The solid state structures of complexes 1 and 3 were evaluated by single crystal X-ray analysis. All complexes were able to hydrolyse the P-O bond of the phosphate monoester in 90% (v/v) DMSO-water medium using 4-nitrophenylphosphate (4-NPP) as the model substrate and the trend in their activity is 1 approximate to 2 > 3. On considering the highly efficient hydrolysis properties, complexes 1-3 were tested as potential therapeutic agents for cancer using HCT116 (human colorectal carcinoma), HepG2 (human hepatocellular carcinoma) and A549 (human non-small lung carcinoma) cells. Complex 2 showed the highest IC50 values for anti-cancer activity towards all three cell lines among the three complexes and complex 3 showed the least activity as observed in the phosphatase activity study. The internucleosomal degradation of DNA during apoptosis can be detected by cell death detection ELISA. DNA fragmentation that leads to cell death was examined and when induced by complex 2 in HepG2 cells a significant level of DNA fragmentation was observed at regular intervals of time.

Benzoylhydrazone Schiff base-ligated three new ONO pincer-type palladium(II) complexes, [(PdL1(PPh3)] (1), [(PdL2(PPh3)] (2), and [(PdL3(PPh3)] (3), were synthesized by the reaction of the respective ligand, N-(2-hydroxybenzylidene)benzohydrazide (HL1), N-(2-hydroxy-3-methoxybenzylidene)benzohydrazide (HL2), or N-(5-bromo-2-hydroxybenzylidene) benzohydrazide (HL3), with Pd(OAc)(2) and PPh3 in methanol and isolated as air-stable reddish-orange crystalline solids in high yields (78%-83%). All three complexes were fully characterized by elemental analysis, Fourier-transform infrared spectroscopy, UV-Visible, H-1 nuclear magnetic resonance (NMR), C-13{H-1} NMR, and P-31{H-1} NMR spectroscopic studies. The molecular structure of all three complexes was established unambiguously by single-crystal X-ray diffraction studies which revealed a distorted square planar geometry of all three complexes. The ONO pincer-type ligands occupied three coordination sites at the palladium, while the fourth site is occupied by the monodentate triphenylphosphine ligand. The catalytic potential of all three complexes was explored in the carbonylative Suzuki coupling of aryl bromides and iodides with arylboronic acids to yield biaryl ketones, using CHCl3 as the source of carbonyl. The reported protocol is convenient and safe as it obviates the use of carbon monoxide (CO) balloons or pressured CO reactors which are otherwise needed for the carbonylation reactions. The methodology has been successfully applied to the synthesis of two antineoplastic drugs, namely, phenstatin and naphthylphenstatin, in good yields (81% and 85%, respectively). Under the optimized reaction conditions, complex 2 exhibited the best catalytic activity in the carbonylative Suzuki couplings. The reported catalysts have wide reaction scope with good functional group tolerance. All catalysts could be retrieved from the reaction after completion and recycled up to three times with insignificant loss in the catalytic activity.

The transfer hydrogenation of ketones catalyzed by transition metal complexes has attracted much attention. A series of ruthenium(11) complexes bearing 2,6-bis(5,6-dialkyl-1,2,4-triazin-3-yl)pyridine ligands (R-BTPs) were synthesized and characterized by NMR analysis and X-ray diffraction. These ruthenium (II) complexes were applied in the transfer hydrogenation of ketones. Their different catalytic activity were attributed to the alkyl arms on the 2,6-bis(5,6-dialkyl-1,2,4-triazin-3-yl)pyridine. As the length of the alkyl arms rising, the catalytic activities of the complex catalysts decreased. By means of 0.4 mol % catalyst RuCl2(PPh3)(3-methylbutyl-BTP) in refluxing 2-propanol, a variety of ketones were reduced to their corresponding alcohols with >95% conversion over a period of 3 h. A series of two new mono- and one binuclear m-nitrato bridged copper(II) complexes [Cu(L)(HL)]ClO4 (1), [Cu(HL)(NO3)(H2O)](2)NO3 center dot H2O (2) and [Cu-2(L-2)(mu-NO3)(2)] (3), with an unsymmetrical NNO donor Schiff base (HL) have been synthesized and characterized by elemental analysis, FTIR, CV, UV-vis and EPR spectroscopy. Their molecular structures were also determined by single crystal X-ray crystallography. In the binuclear complex 3, the Cu center dot center dot center dot Cu distance is 3.494 angstrom. In 1, 2 and 3, the Cu(II) centers have distorted square pyramidal geometry (tau(5) = 0.05-0.17). Evidence of weak pi center dot center dot center dot pi stacking intermolecular interactions along with other non-covalent interactions (hydrogen bonding) was observed by analyzing the respective crystal structures of the complexes. Thus, these hydrogen bonds, pi center dot center dot center dot pi stacking interactions and other weak intermolecular interactions establish in the form of supramolecular architectures a crystalline network environment. The non-covalent interactions were also investigated by employing Hirshfeld Analysis. The room temperature magnetic moments of the mononuclear complexes are less than the spin only values which are indicative of small interactions. Also, significant magnetic interactions were not exhibited by binuclear copper(II) complex 3 in the variable temperature magnetic measurements. The X-band EPR spectra of all three complexes exhibit copper(II) hyperfine structures as well as zero-field splitting which are appropriate for the triplet states of dimers. In complexes 1 and 2, the presence of pseudo dipolar interactions is proposed. Quantum chemical calculations (DFT) were carried out on complexes 1-3 to explore the electronic and spectral properties of these newly synthesized complexes. These complexes show significant antiproliferative and SOD activity. The SOD activity measured in terms of kMcCF is in the range 4.94-12.31 (mol L)(-1)s(-1))x 10(4). (C) 2021 Elsevier Ltd. Here, we report a new and practical strategy for the reductive deuteration of ketones using a Mg/BrCH2CH2Br/D2O system, which affords alpha-deuterated alcohols in good yields and with almost quantitative incorporation of deuterium. The synthetic value of this method has been demonstrated by the easy access to deuterated drugs or drug derivatives. This method may inspire the discovery of other deuterium-containing drugs.

Background: In recent times, coordination complexes of iron in various oxidation states along with variety of ligand systems have been designed and developed for effective treatment of cancer cells without adversely affecting the normal cell and tissues of various organs. Methods: In this study, we have evaluated the mechanism of action of a Fe(II) Schiff base complex in the crop plant Trigonella foenum-graecum L. (Fenugreek) as the screening system by using morphological, cytological, biochemical and molecular approaches. Further functional characterization was performed using MCF-7 cell line and solid tumour model for the assessment of anti-tumour activity of the complex. Results: Our results indicate efficiency of the Fe(II) Schiff base complex in the induction of double strand breaks in DNA. Complex treatment clearly induced cytotoxic and genotoxic damage in Trigonella seedlings. The Fe-complex treatment caused cell cycle arrest via the activation of ATM-ATR kinase mediated DNA damage response pathway with the compromised expression of CDK1, CDK2 and CyclinB1 protein in Trigonella seedlings. In cultured MCF-7 cells, the complex induces cytotoxicity and DNA fragmentation through intracellular ROS generation. Fe-complex treatment inhibited tumour growth in solid tumour model with no additional side effects. Conclusion: The growth inhibitory and cytotoxic effects of the complex result from activation of DNA damage response along with oxidative stress and cell cycle arrest. General significance: Overall, our results have provided comprehensive information on the mechanism of action and efficacy of a Fe(II) Schiff base complex in higher eukaryotic genomes and indicated its future implications as potential therapeutic agent.

A new family of eight heterobimetallic Cu(i)-dppf complexes of general formula [Cu(dppf)L][BF4] with dppf = 1,1 ‘-bis(diphenylphosphino)ferrocene and L representing N,N-, N,O- and N,S-heteroaromatic bidentate ligands have been synthesized and fully characterized by classical analytical, spectroscopic and electrochemical methods. The single crystal structures of [Cu(dppf)(pBI)][BF4] (6), [Cu(dppf)(dpytz)][BF4] (7) and [Cu(dppf)(5-Aphen)][BF4] (8) complexes (where pBI = 2-(2-pyridyl)benzimidazole, dpytz = 3-(2-pyridyl)-5,6-diphenyl-1,2,4-triazine and 5-Aphen = 1,10-phenanthrolin-5-amine) were determined by X-ray diffraction studies. Cytotoxicity of all complexes was evaluated in two human breast adenocarcinoma cell lines (MCF7 and MDAMB231). All the complexes exhibit high cytotoxicity against both human breast cancer cells with IC50 values far lower than those found for the antitumor drug cisplatin in the same cell lines. The IC50 values on primary healthy fibroblasts are of the same order of magnitude as those found for the tumoral cells.

Three asymmetric tridentate acyl hydrazone Schiff base ligands namely L1, L2 and L3 were prepared via condensation of 4-methoxybenzohydrazide with picolinaldehyde, 1-(pyridin-2-yl)ethanone and phenyl(pyridin-2-yl)methanone respectively. Three bio-relevant mononuclear zinc(ii) complexes [Zn(L1)Cl-2]2H(2)O (1), [Zn(L2)Cl-2] (2) and [Zn(L3)Cl-2] (3) were synthesized by treatment of zinc(ii) chloride with the corresponding Schiff base ligands and characterised by the usual physicochemical techniques. The solid state structures of complexes 1 and 3 were evaluated by single crystal X-ray analysis. All complexes were able to hydrolyse the P-O bond of the phosphate monoester in 90% (v/v) DMSO-water medium using 4-nitrophenylphosphate (4-NPP) as the model substrate and the trend in their activity is 1 approximate to 2 > 3. On considering the highly efficient hydrolysis properties, complexes 1-3 were tested as potential therapeutic agents for cancer using HCT116 (human colorectal carcinoma), HepG2 (human hepatocellular carcinoma) and A549 (human non-small lung carcinoma) cells. Complex 2 showed the highest IC50 values for anti-cancer activity towards all three cell lines among the three complexes and complex 3 showed the least activity as observed in the phosphatase activity study. The internucleosomal degradation of DNA during apoptosis can be detected by cell death detection ELISA. DNA fragmentation that leads to cell death was examined and when induced by complex 2 in HepG2 cells a significant level of DNA fragmentation was observed at regular intervals of time.

The carbonylative Suzuki cross-coupling of 2-bromopyridine with various boronic acids to prepare unsymmetrical arylpyridine ketones has been carried out using palladium/N-heterocyclic carbene complexes as catalysts prepared in situ. The selectivity and the rate of these reactions are highly dependent on the conditions, i.e., nature of the palladium catalyst precursor, solvent, temperature and CO pressure. The main side-products arise from direct, non-carbonylative cross-coupling. Under the optimum conditions, arylpyridine ketones are recovered in high yields (60-88%). The antibacterial activities of the corresponding benzimidazole salts 2 were tested against Gram positive and negative bacteria using the agar dilution procedure, and their IC50 values have been determined.
