New Schiff bases [N’-(phenyl(pyridin-2-yl)methylene) isonicotinohydrazide ((LH)-H-1), N-1-(naphthalen-1-yl)-N-2-(phenyl(pyridin-2-yl) methylidene) ethane-1,2-diamine ((LH)-H-2), N-(6-chlorobenzo[d]thiazol-2-yl)-1-phenyl-1-(pyridin-2-yl) methanimine ((LH)-H-3))were synthesized by reaction of 2-benzoylpyridine with different amines (2-amino-6-chlorobenzothiazole, isonicotinohydrazide and N-1-(naphthalen-1-yl)ethane-1,2-diamine) and characterized by H-1-NMR, C-13-NMR, IR mass spectroscopy and elemental analysis. The compounds were assayed by the disc diffusion method for anti-bacterial against five pathogenic bacteria species (Staphylococcus aureus, Micrococcus luteus, Staphylococcus pyogenes, Bacillus sub tilis, and E. coli). All prepared Schiff bases showed good activity compared to positive control (streptomycin), Moreover the (LH)-H-3 showed the highest activity against S. aureus, and M. luteus than the other compounds and streptomycin. In additional molecular docking studies with 3-chymotrypsin-like protease (3CLpro), the essential enzyme for SARS-CoV-2 proliferation. The rest of compounds have shown promising results as 3CLpro inhibitors interacting with the active sites of the enzymes. Finally, DFT ‘s estimated electrostatic molecular potential results were used to illustrate the molecular docking findings. The DFT calculations showed that (LH)-H-3 has the highest dipole moment and electrophilicity index. Interestingly, (LH)-H-2 of the largest energy gap Delta E = 2.49 eV, there are several hydrophilic interactions that could facilitate the binding with the receptors. A series of two new mono- and one binuclear m-nitrato bridged copper(II) complexes [Cu(L)(HL)]ClO4 (1), [Cu(HL)(NO3)(H2O)](2)NO3 center dot H2O (2) and [Cu-2(L-2)(mu-NO3)(2)] (3), with an unsymmetrical NNO donor Schiff base (HL) have been synthesized and characterized by elemental analysis, FTIR, CV, UV-vis and EPR spectroscopy. Their molecular structures were also determined by single crystal X-ray crystallography. In the binuclear complex 3, the Cu center dot center dot center dot Cu distance is 3.494 angstrom. In 1, 2 and 3, the Cu(II) centers have distorted square pyramidal geometry (tau(5) = 0.05-0.17). Evidence of weak pi center dot center dot center dot pi stacking intermolecular interactions along with other non-covalent interactions (hydrogen bonding) was observed by analyzing the respective crystal structures of the complexes. Thus, these hydrogen bonds, pi center dot center dot center dot pi stacking interactions and other weak intermolecular interactions establish in the form of supramolecular architectures a crystalline network environment. The non-covalent interactions were also investigated by employing Hirshfeld Analysis. The room temperature magnetic moments of the mononuclear complexes are less than the spin only values which are indicative of small interactions. Also, significant magnetic interactions were not exhibited by binuclear copper(II) complex 3 in the variable temperature magnetic measurements. The X-band EPR spectra of all three complexes exhibit copper(II) hyperfine structures as well as zero-field splitting which are appropriate for the triplet states of dimers. In complexes 1 and 2, the presence of pseudo dipolar interactions is proposed. Quantum chemical calculations (DFT) were carried out on complexes 1-3 to explore the electronic and spectral properties of these newly synthesized complexes. These complexes show significant antiproliferative and SOD activity. The SOD activity measured in terms of kMcCF is in the range 4.94-12.31 (mol L)(-1)s(-1))x 10(4). (C) 2021 Elsevier Ltd. Anticancer activity of Pd complexes 1-5 with bidentate N-heteroaromatic hydrazone ligands was investigated on human acute monocytic leukemia (THP-1; cells in a suspension) and human mammary adenocarcinoma (MCF-7; two-dimensional layer and three-dimensional spheroid tumor model) cell lines. For the Pd(II) complexes with condensation products of ethyl hydrazainoacetate and quinoline-8-carboxaldehyde (complex 1) and 2-for-mylpyridine (complex 3), for which apoptosis was determined as a mechanism of anticancer activity, further investigation revealed that they arrest the cell cycle in G0/G1 phase, induce generation of reactive oxygen species and inhibit Topoisomerase I in vitro. In silico studies corroborate experimental findings that these complexes show topoisomerase inhibition activity in the micromolar range and indicate binding to a DNA’s minor groove as another potential target. Based on the results obtained by circular dichroism and fluorescence spectroscopy measurements, the most active complexes are suitable to be delivered to a blood stream via human serum albumin.

Flavin-dependent ‘ene’-reductases (EREDs) are highly selective catalysts for the asymmetric reduction of activated alkenes. This function is, however, limited to enones, enoates, and nitroalkenes using the native hydride transfer mechanism. Here we demonstrate that EREDs can reduce vinyl pyridines when irradiated with visible light in the presence of a photoredox catalyst. Experimental evidence suggests the reaction proceeds via a radical mechanism where the vinyl pyridine is reduced to the corresponding neutral benzylic radical in solution. DFT calculations reveal this radical to be dynamically stable, suggesting it is sufficiently long-lived to diffuse into the enzyme active site for stereoselective hydrogen atom transfer. This reduction mechanism is distinct from the native one, highlighting the opportunity to expand the synthetic capabilities of existing enzyme platforms by exploiting new mechanistic models.

Strained compounds are privileged moieties in modern synthesis. In this context, 1-azabicyclo[1.1.0]butanes are appealing structural motifs that can be employed as click reagents or precursors to azetidines. We herein report the first telescoped continuous flow protocol for the generation, lithiation, and electrophilic trapping of 1-azabicyclo[1.1.0]butanes. The flow method allows for exquisite control of the reaction parameters, and the process operates at higher temperatures and safer conditions with respect to batch mode. The efficiency of this intramolecular cyclization/C3-lithiation/electrophilic quenching flow sequence is documented with more than 20 examples.

A simple, and practical oxidative scission of aromatic olefins to carbonyl compounds using O-2 as the sole oxidant with poly(ethylene glycol) dimethyl ether as a benign solvent has been developed. A wide range of monosubstituted, gem-disubstituted, 1,2-disubstituted, trisubstituted and tetrasubstituted aromatic olefins was successfully converted into the corresponding aldehydes and ketones in excellent yields even with gram-scale reaction. Some control experiments were also conducted to support a possible reaction pathway.

Three new mononuclear complexes of the type [M(L)(2)](ClO4)(2), with Ni(II) (1), Cu(II) (2) and Zn(II) (3) ions, have been synthesized and structurally characterized, using the N,N,O donor Schiff base ligand 2-[(phenyl-pyridine-2-yl-methylene)-amino]-ethanol]. An X-ray diffraction study reveals identical crystallization patterns for each of the complexes, in the monoclinic crystal system with the C2/c space group. Interestingly, the synthesized complexes are isostructural in nature. The complexes showed biomimetic phenoxazinone synthase-like activity for the aerial oxidation of o-aminophenol to phenoxazine-2-one, separately in methanol (MeOH) and acetonitrile (ACN) solutions. For each of the complexes, the turnover numbers were found to be higher in MeOH than in ACN. The highest catalytic activity between among the three complexes was found to be for complex 2.
