Steric hindrance in the binding pocket of an alcohol dehydrogenase (ADH) has a great impact on its activity and stereoselectivity simultaneously. Due to the subtle structural difference between two bulky phenyl substituents, the asymmetric synthesis of diaryl alcohols by bioreduction of diaryl ketones is often hindered by the low activity and stereoselectivity of ADHs. To engineer an ADH with practical properties and to investigate the molecular mechanism behind the asymmetric biocatalysis of diaryl ketones, we engineered an ADH from Lactobacillus kefiri (LkADH) to asymmetrically catalyse the reduction of 4-chlorodiphenylketones (CPPK), which are not catalysed by the wild type (WT) enzyme. Mutants seq1-seq5 with gradually increased activity and stereoselectivity were obtained through iterative shrinking mutagenesis. The final mutant seq5 (Y190P/I144V/L199V/E145C/M206F) demonstrated the highest activity and excellent stereoselectivity of >99% ee. Molecular simulation analyses revealed that mutations may enhance the activity by eliminating steric hindrance, inducing a more open binding loop and constructing more noncovalent interactions. The pro-R pose of CPPK with a halogen bond formed a pre-reaction conformation more easily than the pro-S pose, resulting in the high ee of (R)-CPPO in seq5. Moreover, different halogen bonds formed due to the different positions of chlorine substituents, resulting in opposite substrate binding orientation and stereoselectivity. Therefore, the stereoselectivity of seq5 was inverted toward ortho- rather than para-chlorine substituted ketones. These results indicate that the stereocontrol element of LkADH was changed to recognise diaryl ketones after steric hindrance was eliminated. This study provides novel insights into the role of steric hindrance and noncovalent bonds in the determination of the activity and stereoselectivity of enzymes, and presents an approach producing key intermediates of chiral drugs with practical potential.

Asymmetric reduction studies of heteroaryl ketones, including phenyl(pyridin-2-yl)methanone in enantioselective form with biocatalysts are very few, and chiral heteroaryl alcohols have been synthesized generally in the small scale. In this study, seven bacterial strains have been used to produce the (S)-phenyl(pyridin-2-yl)methanol in high enantiomeric excess and yield. Among the tested strains, Lactobacillus paracasei BD101, was found to be the best biocatalyst for the reducing phenyl(pyridin-2-yl)methanone to the (S)-phenyl(pyridin-2-yl)methanol at gram scale. The asymmetric bioreduction conditions were systematically optimized using L. paracasei BD101, which demonstrated excellent enantioselectivity and high level of conversion for the bioreduction reaction. (S)-phenyl(pyridin-2-yl)methanol, which is an analgesic, was produced enantiomerically pure form in the first time on gram scale using a biocatalyst. In total, 5.857 g of (S)-phenyl(pyridin-2-yl)methanol in enantiomerically pure form (>99% enantiomeric excess) was obtained in 52 h with 93% yield using whole cells of L. paracasei BD101. Enantiomerically pure (S)-phenyl (pyridin-2-yl)methanol, which is an analgesic, was first produced in the gram scale using a biocatalyst with excellent ee (>99%) and yield (93%). [GRAPHICS] .

Semicarbazones can exist in two tautomeric forms. In the solid state, they are found in the keto form. This work presents the synthesis, structures and spectroscopic characterization (IR and NMR spectroscopy) of four such compounds, namely the neutral molecule 4-phenyl-1-[phenyl(pyridin-2-yl)methylidene] semicarbazide, C19H16N4O, (I), abbreviated as HBzPyS, and three different hydrated salts, namely the chloride dihydrate, C19H17N4O+center dot Cl-center dot 2H(2)O, (II), the nitrate dihydrate, C19H17N4O+center dot NO3-center dot 2H(2)O, (III), and the thiocyanate 2.5-hydrate, C19H17N4O+center dot SCN-center dot 2.5H(2)O, (IV), of 2-[phenyl({[(phenylcarbamoyl) amino]imino})methyl]pyridinium, abbreviated as [H(2)BzPyS](+)center dot X-center dot nH(2)O, with X = Cl- and n = 2 for (II), X= NO3- and n = 2 for (III), and X = SCN- and n = 2.5 for (IV), showing the influence of the anionic form in the intermolecular interactions. Water molecules and counter-ions (chloride or nitrate) are involved in the formation of a two-dimensional arrangement by the establishment of hydrogen bonds with the N-H groups of the cation, stabilizing the E isomers in the solid state. The neutral HBzPyS molecule crystallized as the E isomer due to the existence of weak pi-pi interactions between pairs of molecules. The calculated IR spectrum of the hydrated [H(2)BzPyS](+) cation is in good agreement with the experimental results.

A series of bidendate pyridine-functionalized metal N-heterocyclic carbene (NHC) complexes with various wingtip substituents (R = methyl, phenyl and tert-butyl) had been prepared from silver oxide-mediated transmetalation method. The basic backbone of the NHC ligands were prepared in two step-wise reactions: reduction of the commercially available 2-benzoylpyridine, followed by halogenation to yield 2-(chloro(phenyl) methyl) pyridine. Subsequent reactions with different imidazoles gave the desired imidazolium salts (or NHC precursors) with different R groups at the N wingtip (R = Me, Ph, t-Bu). Transmetalation of the imidazolium salts into platinum NHC complexes were achieved by using Ag2O mediated method with Pt(cod)Cl-2 as the transmetalating agent. On the other hand, nickel NHC complexes were obtained via one pot synthesis method with nickel chloride as the transmetalating agent and reflux in the presence of a base. Following recrystallisation, the molecular structure of Pt NHC complex (+/-)-5a had been successfully elucidated via single crystal X ray diffraction (XRD) analysis. The six-membered ring of the platinum NHC complexes adopted a boat conformation with the phenyl ring arranged at the axial position. Meanwhile, two unexpected nickel complexes, i.e. a simple nickel coordination complex and a nickelate complex, were obtained and studied using XRD. These Pt NHC complexes and unexpected Ni complexes were evaluated for their in vitro antimicrobial and anticancer activities against a panel of pathogenic microorganisms and three selected human cancer cell lines, including breast (MCF7), colon (HCT116) and oral (H103) cancer cells. Generally, the Pt NHC complexes displayed enhanced antimicrobial activities upon coordination to metals, as compared to the corresponding imidazolium salts i.e. NHC precursors, with minimum inhibitory concentration (MIC) values in micromolar (mM) range. A couple of platinum NHC complexes synthesized exhibited antimicrobial activities with MIC as low as 2 mM, which are comparable to silver NHC complexes with renowned antimicrobial profiles. Similarly, upon coordination to the metals, the cytotoxic effects of the platinum NHC complexes increased significantly as compared to the imidazolium salts. Among all, platinum NHC complex (+/-)-5c displayed significant cytotoxicities towards the cancer cells with IC50 values that are two to three times lower than that of the anticancer drug cisplatin. The electrochemical behavior of a series of the metal complexes [Fe(III), Ni(II), Ru(III),Pd(II) and Hf(IV)]derived from the Schiff base ligand(E)-5-((phenyl(-pyridin-2-yl) methylene) amino) pyrimidine-2,4(1H, 3H)-dione(H2L) which was previously prepared from condensation of 5-aminouracil and 2-benzoylpyridine was studied by using cyclic voltammetric technique. The Schiff base ligand H2L and its metal complexes exhibited quasi- reversible oxidation- reduction with three electron transfer and it was suggested that their reactions on the platinum surface electrode are not purely diffusion controlled but also under adsorption control. The redox reactive sites of the H2L and its complexes were located via the geometry optimization and frequency calculations using density functional theory (DFT) at the B3LYP/LanL2DZ level of theory. In addition, the Schiff base ligand and its metal complexes exhibit fluorescent properties. The antimicrobial activity of the Schiff base ligand H2L and its metal complexes was tested against Gram-positive bacteria (Staphylococcus aureus and Bacillus cereus(,Gram-negative bacteria (E. coli and Pseudomonas aeruginosa) and fungal strain (Aspergillus niger) by using agar well-diffusion method. Structure-activity relationships for a series of small-molecule thiophenes resulted in potent and selective antagonism of human Complement C3a receptor. Three new mononuclear complexes of the type [M(L)(2)](ClO4)(2), with Ni(II) (1), Cu(II) (2) and Zn(II) (3) ions, have been synthesized and structurally characterized, using the N,N,O donor Schiff base ligand 2-[(phenyl-pyridine-2-yl-methylene)-amino]-ethanol]. An X-ray diffraction study reveals identical crystallization patterns for each of the complexes, in the monoclinic crystal system with the C2/c space group. Interestingly, the synthesized complexes are isostructural in nature. The complexes showed biomimetic phenoxazinone synthase-like activity for the aerial oxidation of o-aminophenol to phenoxazine-2-one, separately in methanol (MeOH) and acetonitrile (ACN) solutions. For each of the complexes, the turnover numbers were found to be higher in MeOH than in ACN. The highest catalytic activity between among the three complexes was found to be for complex 2.
