Ru(II)-p-cymene complexes (1-3) containing picolyl based pseudo-acylthiourea ligands (L-1-L-3) were synthesized and characterized. The crystallographic study confirmed the molecular structures of all the ligands (L-1-L-3) and complex 3. The catalytic activity of the complexes was tested mainly towards TH of carbonyl compounds and nitroarenes. The influence of steric and electronic effects of the ligands on the chemoselectivity and reactivity were reported. The catalytic activity was enhanced and chemoselectivity was switched after tuning the ligands in the catalysts, compared to their corresponding unmodified Ru(II)-p-cymene complexes. The catalysis was extended to a broad range of substrates including some challenging systems like furfural, benzoylpyridine, benzoquinone, chromanone, etc. The strategy of tuning the bifunctional ligands in the catalysts for effective and selective catalysis worked nicely. Further, the catalysis was extended to one pot synthesis of 3-isopropoxyindole from 2-nitrocinnamaldehyde, the first synthetic route similar to Baeyer Emmerling indole synthesis. A new aroylhydrazone ligand, 2-benzoylpyridine-4-methoxybenzhydrazone (BPMBH). and its two cobalt(III) complexes, [Co(BPMB)(2)]NO3 center dot 1.5H(2)O (1) and [Co(BPMB)(2)]Cl center dot 2H(2)O (2) have been synthesized and structurally characterized. The synthesized compounds are physico-chemically characterized by CHN analysis, molar conductivity, magnetic susceptibility measurements and spectroscopic techniques like MS, IR, UV and NMR. The high molar conductivity values of the complexes confirmed the presence of counter ions in the lattice of the cationic complexes. The molecular structure of the aroylhydrazone and its complexes have been resolved using single crystal XRD studies. In the solid state, the aroylhydrazone exists in the amido form as evident from the IR and XRD studies. The tridentate nature of the NNO donor aroylhydrazone was also confirmed from the IR spectral studies. XRD studies reveal that in all complexes, the tridentate aroylhydrazone coordinates to the Co(III) center via pyridine nitrogen, azomethine nitrogen and iminolate oxygen and found to possess distorted octahedral geometry. The complexes are bis-ligated cationic complexes in which chloride and nitrate ions act as counter ions. The antibacterial and antifungal activities of the aroylhydrazone and its Co(III) complexes have been screened against bacterial species Staphylococcus aureus, Enterobacter aerogenes, Bacillus subtilis, Streptococcus pyogenes, Klebsiella pneumonia, Salmonella typhi and Escherichia coli. The antifungal activity was also checked using fungi Aspergillus niger, Penicillium chrysogenum and Rhizopus oryzae. From the activity data we can infer that the metal complexes have better biological activity than the metal free ligand against the bacterial and fungal species which were tested. Green and sustainable chemistry approaches necessitate an ongoing investigation towards new, environmentally benign and selective catalysts. We have thus prepared a new benzothiazole-scaffolded hydrazone Schiff base ligand L and coordinated it with copper(II) ions leading to five different complexes, the form of which in the solid state is counterion-dependent. Nitrate, chloride and trifiate anions lead to monometallic species, whereas utilization of sulfates and tetrafluoroborates is responsible for the formation of bimetallic assemblies. The catalytic efficiency of synthesized compounds was shown in Cu-TEMPO (2,2,6,6-tetramethyl-l-piperidinoxyl) aerobic oxidation of activated alcohols resulting in good to excellent conversions and 100% selectivity to form the corresponding aldehydes. Considering the experimental results and Density Functional Theory calculations we propose two most plausible reaction mechanisms that further corroborate the enhanced activity of bimetallic species. It appears that the tridentate character of the ligand sacrifices some of the overall catalytic efficiency for the selectivity of the process. Aqueous reaction medium, low catalyst loading, air as oxidant and exclusive oxidative selectivity render these Cu ll complexes promising candidates for further improvement. (C) 2020 Elsevier B.V. Background: In recent times, coordination complexes of iron in various oxidation states along with variety of ligand systems have been designed and developed for effective treatment of cancer cells without adversely affecting the normal cell and tissues of various organs. Methods: In this study, we have evaluated the mechanism of action of a Fe(II) Schiff base complex in the crop plant Trigonella foenum-graecum L. (Fenugreek) as the screening system by using morphological, cytological, biochemical and molecular approaches. Further functional characterization was performed using MCF-7 cell line and solid tumour model for the assessment of anti-tumour activity of the complex. Results: Our results indicate efficiency of the Fe(II) Schiff base complex in the induction of double strand breaks in DNA. Complex treatment clearly induced cytotoxic and genotoxic damage in Trigonella seedlings. The Fe-complex treatment caused cell cycle arrest via the activation of ATM-ATR kinase mediated DNA damage response pathway with the compromised expression of CDK1, CDK2 and CyclinB1 protein in Trigonella seedlings. In cultured MCF-7 cells, the complex induces cytotoxicity and DNA fragmentation through intracellular ROS generation. Fe-complex treatment inhibited tumour growth in solid tumour model with no additional side effects. Conclusion: The growth inhibitory and cytotoxic effects of the complex result from activation of DNA damage response along with oxidative stress and cell cycle arrest. General significance: Overall, our results have provided comprehensive information on the mechanism of action and efficacy of a Fe(II) Schiff base complex in higher eukaryotic genomes and indicated its future implications as potential therapeutic agent.

A highly direct asymmetric reductive amination of a variety of ketone substrates, including 2-acetyl-6-substituted pyridines, beta-keto esters, beta-keto amides, and 1-(6-methylpyridin-2yl)propan-2-one, has been disclosed for the first time (94.6% to >99.9% ee). With ammonium trifluoroacetate as the nitrogen source, various chiral corresponding primary amines were prepared in excellent enantioselectivity and conversion in the presence of a commercially available and inexpensive chiral catalyst, Ru(OAc)(2){(S)-binap}, under 0.8 MPa of hydrogen gas pressure.

Local lipid variations in tissues are readily revealed with mass spectrometry imaging (MSI) methods, and the resulting lipid distributions serve as bioanalytical signatures to reveal cell- or tissue-specific lipids. Comprehensive MSI lipid mapping requires measurements in both ion polarities. Additionally, structural lipid characterization is necessary to link the lipid structure to lipid function. Whereas some structural elements of lipids are readily derived from high-resolution mass spectrometry (MS) and tandem-MS (MSn), the localization of C=C double bonds (DBs) requires specialized fragmentation and/or functionalization methods. In this work, we identify a multifunctional matrix-assisted laser desorption/ionization (MALDI) matrix for spatially resolved lipidomics investigations that reacts with lipids in Paterno-Biichi (PB) reactions during laser irradiation facilitating DB-position assignment and allows dual-polarity high-resolution MALDI-MSI and MALDI (MSI)-I-2 studies. By screening 12 compounds for improved ionization efficiency in positive-/negative-ion mode and the functionalization yield compared to the previously introduced reactive MALDI matrix benzophenone, 2-benzoylpyridine (BzPy) is identified as the best candidate. The new matrix enables DB localization of authentic standards belonging to 12 lipid classes and helps to assign 133/58 lipid features in positive-/negative-ion mode from mouse cerebellum tissue. The analytical capabilities of BzPy as a multifunctional MALDI-MSI matrix are demonstrated by imaging endogenous and PB-functionalized lipids in mouse kidney sections with 7 mu m lateral resolution in both ion modes. Tracking diagnostic lipid DB-position fragment ions in mouse pancreatic tissue with down to 10 mu m pixel size allows us to identify the islets of Langerhans associated with lipid isomer upregulation and depletion.

N-aminopyridyl ketone salts were reacted with formamide to yield heteroaromatic pyrido[2,1-f][1,2,4] triazinium salts. Upon storage of these products in the presence of water, formation of covalent hydrates have been observed. Reaction of the same starting compound with urethane yielded 3-chloropyrido[2,1-f][1,2,4]triazinium salt which readily reacted with secondary amines to afford 3-amino derivatives. An analogous ring closure reaction of 2-formylaminomethyl- and formaminobenzylpyridine allowed the synthesis of the partially reduced 3,4-dihydropyrido[2,1-f][1,2,4]triazinium compounds. Three imidazole hydrazone compounds, namely 2-(4-nitro-1H-imidazol-1-yl)-N’-[1-(pyridin-2-yl)ethylidene]acetohydrazide, C12H12N6O3, (1), 2-(2-nitro-1H-imidazol-1-yl)-N’-[1-(pyridin-2-yl)ethylidene]acetohydrazide, C12H12N6O3, (2), and 2-(2-nitro-1H-imidazol-1-yl)-N’-[(phenyl)(pyridin-2-yl)methylidene]acetohydrazide, C17H14N6O3, (3), were obtained and fully characterized, including their crystal structure determinations. While all the compounds proved not to be cytotoxic to J774.A1 macrophage cells, (1) and (3) exhibited activity against Leishmania chagasi, whereas (2) was revealed to be inactive. Since both (1) and (3) exhibited antileishmanial effects, while (2) was devoid of activity, the presence of the acetyl or benzoyl groups was possibly not a determining factor in the observed antiprotozoal activity. In contrast, since (1) and (3) are 4-nitroimidazole derivatives and (2) is a 2-nitroimidazole-derived compound, the presence of the 4-nitro group probably favours antileishmanial activity over the 2-nitro group. The results suggested that further investigations on compounds (1) and (3) as bioreducible antileishmanial prodrug candidates are called for.

A new metal-free procedure allowing Csp(3)-N bond formation through coupling of pyridotriazoles and weakly nucleophilic anilines has been developed. This sustainable reaction shows high tolerance towards functional groups (ketones, free alcohols) leading to 2-picolylamine derivatives. The key to our success is the use of a catalytic amount of TBAB and water as a co-solvent leading to the formation of pyridylalkylamine derivatives. As this coupling tolerates the presence of Csp(2)-Br bond on both partners of the reaction, we performed a sequential one-pot reaction between functionalized triazolopyridines and anilines followed by a second coupling with N-tosylhydrazones leading to the formation of Csp(3)-N and Csp(2)-Csp(2) bonds.
