Here we present a comprehensive study on the [3+2]-cycloaddition of thiocarbonyl ylides with a wide variety of alkenes and alkynes. The obtained dihydro- and tetrahydrothiophene products serve as exceptionally versatile intermediates providing access to thiophenes, dienes, dendralenes, and vic-quarternary carbon centers. The use of high-pressure conditions enables thermally unstable, sterically encumbered or moderately reactive substrates to undergo the cycloaddition under mild conditions, thereby increasing the yield by up to 58%. In addition, we showcase its utility by the formal syntheses of the pharmaceuticals NGB 4420 and tenilapine.

A three-dimensional database search has been applied to design a series of endo- and exo-3-(pyridin-3-yl)bicyclo[2.2.1]heptan-2-amines as nicotinic receptor ligands. The synthesized compounds were tested in radioligand binding assay on rat cortex against [H-3]-cytisine and [3H]-methyllycaconitine to measure their affinity for alpha 4 beta 2* and alpha 7* nicotinic receptors. The new derivatives showed some preference for the alpha 4 beta 2* over the alpha 7* subtype, with their affinity being dependent on the endo/exo isomerism and on the methylation degree of the basic nitrogen. The endo primary amines displayed the lowest K-i values on both receptor subtypes. Selected compounds (1a, 2a, 3a, and 6a) were tested on heterologously expressed alpha 4 beta 2, alpha 7, and alpha 3 beta 2 receptors and on SHSY-5Y cells. Compounds 1a and 2a showed alpha 4 beta 2 antagonistic properties while behaved as full agonists on recombinant alpha 7 and on SHSYSY cells. On the alpha 3 beta 2 subtype, only the chloro derivative 2a showed full agonist activity and submicromolar potency (EC50 = 0.43 mu M). The primary amines described here represent new chemotypes for the alpha 7 and alpha 3* receptor subtypes.

A study was carried out to determine systematically the key aroma-active compounds of Langyatai Baijiu with Jian flavour (LBJF) using sensory omics analysis (SOA). A total of 56 odorants were screened out using gas chromatography-olfactometry-mass spectrometry (GC-O-MS)/Osme analysis. Among them, 15 aroma-active components were first identified. After quantitation, 30 odorants had odour activity values (OAVs) > 1.0 in LBJF. Recombinant and omission experiments proved that the esters, alcohols, acids, especially ethyl hexanoate, gamma psi psi-nonalactone, and dimethyl trisulfide, were critical to the flavour of LBJF. The basic and commercial liquors had obvious differences in the skeleton compositions of esters and acids. This study uncovers the characteristics of Jian flavour Baijiu (JFB) and provides a scientific basis for the quality control of JFB, which is helpful for the development of Chinese Baijiu flavour styles.

A naphthalimide-pyridoxal conjugate 1 has been designed and synthesized. Compound 1 forms a stable greenish yellow colored gel in DMSO:H2O (8:1 v/v). Rheological study reveals that the gel is mechanically strong (G> G) over a wide range of applied strains. The morphology of the gel as determined by FESEM shows a highly cross-linked fibrous network. The gel is anion-responsive and is selectively transformed into a sol with a color change from greenish yellow to deep blue only in the presence of F- among other anions. In CH3CN, compound 1 was also sensitive to basic anions such as F- and AcO- ions. In solution, F- was differentiated from AcO- through a color change. While the yellow colored solution of 1 in acetonitrile was changed into deep blue in the presence of F-, AcO- ions gave a faint blue coloration. A similar colorimetric differentiation of F- from AcO- has been possible in CH3CN by a reusable Schiff base-linked Merrifield resin 1a or 1b.

Herein, we realized the simultaneous online detection of six tobacco alkaloids (TM) by in-tube solid-phase microextraction (In-tube SPME) coupled with mass spectrometry by a rapid, sensitive, and matrix effect-free method requiring no chromatographic separation and only minimal sample pre-treatment. A poly (N-isopropylacrylamide-co-divinylbenzene-co-N, N’-methylenediacrylamide) [Poly (NIPAAm-co-DVB-co-MBAA)] monolithic column was designed according to the chemical structures of selected TM and used as an extraction medium engaging in hydrophobic, pi-pi, and hydrogen bonding interactions with analytes, allowing them to be effectively extracted. A number of important parameters were systematically optimized to achieve maximal extraction efficiency. The ion intensity of the TM signals obtained by in-tube SPME-MS were higher than the direct MS mode by about 400 folds with the signal-to-noise ratio improved by 2-7 folds. The detection limits of the six TM were determined as 1.99-4.06 ng g(-1), with good linearity with correlation coefficients exceeding 0.99 obtained under optimal extraction conditions. Besides, TA recoveries in cigarette tobacco spiked at three concentration levels were in the range of 76.4-100.2%, and the corresponding RSDs (n = 5) were obtained as 4.32-7.16%. The extraction performance of the poly (NIPAAm-co-DVB-co-MBAA) monolithic column was well reproducible, with intra- or inter-day precision RSDs determined not to exceed 7.38%. Finally, no marked matrix effects were observed when the developed method was applied to the analysis of both high-abundance and trace level TM in practical samples, and the above technique was therefore concluded to be well suited for the detection of TAs in cigarette tobacco or other products.

Pyrido[1,2-a]indoles are known for medicinally and pharmaceutically important compounds; however, the efficient synthetic routes are scarce in the literature. We report herein a convenient and efficient route to synthesize these molecules through indium-catalyzed transannulation of pyridotriazoles with arenes. Herein we report, a mononuclear, highly fluorescent zinc(ii) complex Zn(L)(N-3)(H2O) (1) that was prepared by an easy one pot method, in which the tridentate Schiff base ligand (E)-4-((2-(dimethylamino)ethylimino)methyl)-5-(hydroxymethyl)-2-methylpyridin-3-ol (HL) was generated in situ. The compound is characterized by various spectroscopic techniques, and its structure was determined by single crystal X-ray diffraction studies. DFT calculations were used to understand the electronic structures of the ligand and the complex, and TD-DFT calculations were performed to interpret the nature of the electronic transitions observed in their UV-vis spectra. In the complex, Zn(II) is found to be penta-coordinated with one azide ligand, an aqua ligand and a monoanionic tridentate N,N,O-donor ligand. In an aqueous methanol (1:9 by volume) solution, at the physiological pH (0.01 M Tris-HCl buffer, pH 7.4), compound 1 exhibits an intense greenish blue fluorescence (lambda(ex) 390 nm, lambda(em) 462 nm), whose intensity is about 17-fold stronger than that of the free ligand. Compound 1 is found to show significant DNA binding activity. The pyridoxal appended tridentate ligand can be used for the bio-imaging of Zn(II).

Herein, we report a graphene oxide (GO) catalyzed condensation of non-activated esters and amines, that can enable diverse amides to be synthesized from abundant ethyl esters forming only volatile alcohol as a by-product. GO accelerates ester to amide conversion in the absence of any additives, unlike other catalysts. A wide range of ester and amine substrates are screened to yield the respective amides in good to excellent yields. The improved catalytic activity can be ascribed to the oxygenated functionalities present on the graphene oxide surface which forms H-bonding with the reactants accelerating the reaction. Improved yields and a wide range of functional group tolerance are some of the important features of the developed protocol.

Oxidized products have become increasingly valuable as building blocks for a wide variety of different processes and fine chemistry, especially in the benzylic position. We report herein a sustainable protocol for this transformation through C-H functionalization and is performed using electrochemistry as the main power source and tert-butyl hydroperoxide as the radical source for the C-H abstraction. The temperature conditions reported here do not increase above 50 degrees C and use an aqueous-based medium. A broad substrate scope is explored, along with bioactive molecules, to give comparable and increased product yields when compared to prior reported literature without the use of electrochemistry.
