Mixed ligand nickel(II) and ruthenium(II) complexes were synthesized from pyridoxal-N-allyl-thiosemicarbazone hydrochloride and triphenylphosphine. The structures of the complexes have been characterized by elemental analysis, IR, H-1 and P-31 NMR, conductivity, magnetic moment measurements and single-crystal X-ray diffraction technique. Based on X-ray crystallographic studies, a square-planar structure has been proposed for the Ni(II) complex, in which the thiosemicarbazone ligand acts as dianionic tridentate ONS ligand. In the case of the Ru(II) complex, the thiosemicarbazone is coordinated to metal atom as a monoanionic bidentate NS donor ligand in an octahedral geometry. Antioxidant activities of the ligand and its metal complexes were calculated as their trolox equivalent antioxidant capacities (TEAC) by CUPRAC method and DPPH assay. Both the ligand and its metal complexes were found to be antioxidant and are much more antioxidant at least 2.1 times than trolox, even, ligand is 3.5 times greater than that of trolox according to CUPRAC. A linear correlation (correlation coefficient R-2 = 0.9997) appeared between the obtained TEAC values by the two antioxidant assays.

The Rotimer, a rotifer-specific biopolymer, is an exogenic bioactive exudate secreted by different monogonant species (e.g. Euchlanis dilatata or Lecane bulla). The production of this viscoelastic biomolecule is induced by different micro-particles, thereby forming a special Rotimer-Inductor Conglomerate (RIC) in a web format. In this case, the water insoluble Carmine crystals, filtered to size (max. diameter was 50 mu m), functioned as an inductor. The RIC production is an adequate empirical indicator to follow up this filamentous biopolymer secretion experientially; moreover, this procedure is very sensitive to the environmental factors (temperature, pH, metals and possible natural pollutant agents). The above mentioned species show completely different reactions to these factors, except to the presence of calcium and to the modulating effects of different drugs. One of the novelties of this work is that the Rotimer secretion and consequently, the RIC-formation is a mutually obligatory and evolutionary calcium-dependent process in the concerned monogonants. This in vivo procedure needs calcium, both for the physiology of animals and for fiber formation, particularly in the latter case. The conglomerate covered area (%) and the detection of the longest filament (mm) of the given RIC were the generally and simultaneously applied methods in the current modulating experiments. Exploring the regulatory (e.g. calciumdependency) and stimulating (e.g. Lucidril effect) possibilities of biopolymer secretion are the basis for optimizing the RIC-production capacities of these micro-metazoans.

Several palladium(ii) and platinum(ii) complexes (1-20) of general formula [M(L-n)(X)(Y)] [M = Pd, X = Y = Cl (1-Cl-4-Cl), X = Y = OAc (1-OAc-4-OAc); M = Pt: X = Y = Cl (5-8); M = Pd, X = Cl, Y = CH3 (9-12); M = Pt, X = Cl, Y = CH3 (13-16) or X = Y = CH3 (17-20); n = 1-4] have been synthesized by reaction of different Pd(ii) and Pt(ii) derivatives with various 3-substituted 1-(2-pyridyl)-imidazo[1,5-a]pyridines; i.e.L-n = 1-(2-pyridyl)-3-arylimidazo[1,5-a]pyridine (aryl = Phenyl, L-1; 2-o-Tolyl, L-2; Mesityl, L-3) and 1-(2-pyridyl)-3-benzylimidazo[1,5-a]pyridine (L-4). The X-ray structural analysis performed on two of Pd(ii) and three Pt(ii) complexes, namely [Pd(L-2)(CH3)Cl] (10), [Pd(L-3)(CH3)Cl] (11) and [Pt(L-1)Cl-2] (5), [Pt(L-4)Cl-2] (8), [Pt(L-2)(CH3)Cl] (14) confirmed the spectroscopic and analytical data. Finally DFT studies unveiled the structural reasons behind the inertia of the synthesised compounds toward metalation, identified as the higher angle steric strain in comparison with the analogous bipyridine complexes.

Schiff bases (SB) obtained from S-methyl dithiocarbazate and aromatic aldehydes: salicylaldehyde (H2L1), ovanillin (H2L2), pyridoxal (H2L3) and 2,6-diformyl-4-methylphenol (H3L4), and their corresponding Zn(II)complexes (1-4), are synthesized. All compounds are characterized by elemental analyses, infrared, UV-Vis, nuclear magnetic resonance spectroscopy and mass spectrometry. The structures of H2L2 and [Zn-2(L-1)(2)(H2O) (DMF)] (1a) (DMF = dimethylformamide) are solved by single crystal X-ray diffraction. The SB coordinates the metal center through the Ophenolate, Nimine and Sthiolate atoms. The radical scavenging activity is tested using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, with all ligand precursors showing IC50 values similar to 40 mu M. Cytotoxicity studies with several tumor cell lines (PC-3, MCF-7 and Caco-2) as well as a non-tumoral cell line (NHDF) are reported. Interestingly, 1 has relevant and selective antiproliferative effect against Caco-2 cells (IC50 = 9.1 mu M). Their antimicrobial activity is evaluated in five bacterial strains (Klebsiella pneumoniae, Acinetobacter baumannii, Listeria monocytogenes, Pseudomonas aeruginosa and Staphylococcus aureus) and two yeast strains (Candida albicans and Candida tropicalis) with some compounds showing bacteriostatic and fungicidal activity. The minimal inhibitory concentration (MIC90) of HnL against Mycobacterium tuberculosis is also reported, with H2L2 and H3L4 showing very high activity (MIC90 < 0.6 mu g/mL). The ability of the compounds to bind bovine serum albumin (BSA) and DNA is evaluated for H3L4 and [Zn-2(L-4)(CH3COO)] (4), both showing high binding constants to BSA (ca. 106 M 1) and ability to bind DNA. Overall, the reported compounds show relevant antitumor and antimicrobial properties, our data indicating they may be promising compounds in several fields of medicinal chemistry. Background De novo lipogenesis (DNL) is the primary metabolic pathway synthesizing fatty acids from carbohydrates, protein, or alcohol. Our aim was to examine associations of in vivo levels of selected fatty acids (16:0, 16:1n7, 18:0, 18:1n9) in DNL with incidence of type 2 diabetes (T2D). Methods and findings Seventeen cohorts from 12 countries (7 from Europe, 7 from the United States, 1 from Australia, 1 from Taiwan; baseline years = 1970-1973 to 2006-2010) conducted harmonized individual-level analyses of associations of DNL-related fatty acids with incident T2D. In total, we evaluated 65,225 participants (mean ages = 52.3-75.5 years; % women = 20.4%62.3% in 12 cohorts recruiting both sexes) and 15,383 incident cases of T2D over the 9-year follow-up on average. Cohort-specific association of each of 16:0, 16:1n7, 18:0, and 18:1n9 with incident T2D was estimated, adjusted for demographic factors, socioeconomic characteristics, alcohol, smoking, physical activity, dyslipidemia, hypertension, menopausal status, and adiposity. Cohort-specific associations were meta-analyzed with an inverse-varianceweighted approach. Each of the 4 fatty acids positively related to incident T2D. Relative risks (RRs) per cohort-specific range between midpoints of the top and bottom quintiles of fatty acid concentrations were 1.53 (1.41-1.66; p< 0.001) for 16:0, 1.40 (1.33-1.48; p< 0.001) for 16:1n-7, 1.14 (1.05-1.22; p = 0.001) for 18:0, and 1.16 (1.07-1.25; p< 0.001) for 18:1n9. Heterogeneity was seen across cohorts (I-2 = 51.1%-73.1% for each fatty acid) but not explained by lipid fractions and global geographical regions. Further adjusted for triglycerides (and 16:0 when appropriate) to evaluate associations independent of overall DNL, the associations remained significant for 16:0, 16:1n7, and 18:0 but were attenuated for 18:1n9 (RR = 1.03, 95% confidence interval (CI) = 0.94-1.13). These findings had limitations in potential reverse causation and residual confounding by imprecisely measured or unmeasured factors. Conclusions Concentrations of fatty acids in the DNL were positively associated with T2D incidence. Our findings support further work to investigate a possible role of DNL and individual fatty acids in the development of T2D. In this work, it is described the design of twenty-four heterocyclic amine-azachalcones compounds through molecular hybridization of chalcone scaffold and fragments of isoniazid, fluoroquinolones, and linezolid with antituberculosis potential. The new compounds were synthesized via Claisen-Schmidt condensation, providing yields of 36-95%. Fifteen compounds showed antituberculosis activity against Mycobacterium tuberculosis H37Rv strain. Two amine-azachalcones 15 and 17 showed relevant biological activity with minimum inhibitory concentration (MIC) values of 6.62 and 4.85 mu M, respectively. Compound 12 showed the best profile of antitubercular activity with MIC = 9.54 mu M and selectivity index (SI) = 9.33. It was found that morpholine group is important to increase potency of antimycobacterial activity but also to add some toxicity to the chalcone molecular framework. The results described herein would be a guide in the designing of novel and optimized antitubercular derivatives based on the chalcone scaffold.

In this study, the chemical and sensory profiles of 42 different nonalcoholic beer (NAB) brands/styles already on the global market and produced through several different brewing techniques were evaluated. A trained panel (i.e., 11 panelists) performed standard-driven descriptive and check-all-that-apply analyses in triplicate to sensorially characterize the aroma and taste/mouthfeel profiles of 42 commercial NABs, a commercial soda, and a commercial seltzer water (n = 44). These beers were also chemically deconstructed using several different analytical techniques targeting volatile and nonvolatile compounds. Consumer analysis (n = 129) was then performed to evaluate the Northern Californian consumer hedonic liking of a selection (n = 12) of these NAB brands. These results provide direction to brewers and/or beverage producers on which techniques they should explore to develop desirable NAB offerings and suggest chemical targets that are indicators of specific flavor qualities and/or preference for American consumers.

A new aroylhydrazone ligand, 2-benzoylpyridine-4-methoxybenzhydrazone (BPMBH). and its two cobalt(III) complexes, [Co(BPMB)(2)]NO3 center dot 1.5H(2)O (1) and [Co(BPMB)(2)]Cl center dot 2H(2)O (2) have been synthesized and structurally characterized. The synthesized compounds are physico-chemically characterized by CHN analysis, molar conductivity, magnetic susceptibility measurements and spectroscopic techniques like MS, IR, UV and NMR. The high molar conductivity values of the complexes confirmed the presence of counter ions in the lattice of the cationic complexes. The molecular structure of the aroylhydrazone and its complexes have been resolved using single crystal XRD studies. In the solid state, the aroylhydrazone exists in the amido form as evident from the IR and XRD studies. The tridentate nature of the NNO donor aroylhydrazone was also confirmed from the IR spectral studies. XRD studies reveal that in all complexes, the tridentate aroylhydrazone coordinates to the Co(III) center via pyridine nitrogen, azomethine nitrogen and iminolate oxygen and found to possess distorted octahedral geometry. The complexes are bis-ligated cationic complexes in which chloride and nitrate ions act as counter ions. The antibacterial and antifungal activities of the aroylhydrazone and its Co(III) complexes have been screened against bacterial species Staphylococcus aureus, Enterobacter aerogenes, Bacillus subtilis, Streptococcus pyogenes, Klebsiella pneumonia, Salmonella typhi and Escherichia coli. The antifungal activity was also checked using fungi Aspergillus niger, Penicillium chrysogenum and Rhizopus oryzae. From the activity data we can infer that the metal complexes have better biological activity than the metal free ligand against the bacterial and fungal species which were tested. A new Schiff base receptor (2-amino-3-(((3-hydroxy-5-(hydroxymethyl)-2-methylpyridin-4-yl)methylene) amino)maleonitrile (GAL)) has been synthesized and such diaminomaleonitrile-based molecular framework is observed to be water soluble. GAL possesses both colorimetric and off-on fluorescent response in the presence of ClO-. The response time has been controlled within 6 min. The limit of detection (LOD) has been calculated to be 47.5 nM. The addition of Cu2+ can only induce clear color evolution from pale to deep yellow (LOD: 0.22 mu M) and no fluorescence changes are found. Moreover, its reliability and practicality are verified via the determination of ClO- in spiked samples of tap water and pond water. Ru(II)-p-cymene complexes (1-3) containing picolyl based pseudo-acylthiourea ligands (L-1-L-3) were synthesized and characterized. The crystallographic study confirmed the molecular structures of all the ligands (L-1-L-3) and complex 3. The catalytic activity of the complexes was tested mainly towards TH of carbonyl compounds and nitroarenes. The influence of steric and electronic effects of the ligands on the chemoselectivity and reactivity were reported. The catalytic activity was enhanced and chemoselectivity was switched after tuning the ligands in the catalysts, compared to their corresponding unmodified Ru(II)-p-cymene complexes. The catalysis was extended to a broad range of substrates including some challenging systems like furfural, benzoylpyridine, benzoquinone, chromanone, etc. The strategy of tuning the bifunctional ligands in the catalysts for effective and selective catalysis worked nicely. Further, the catalysis was extended to one pot synthesis of 3-isopropoxyindole from 2-nitrocinnamaldehyde, the first synthetic route similar to Baeyer Emmerling indole synthesis.
