Background: Bis(indolyl)methane derivatives are widely found in nature with a broad range of biological and pharmacological activities. The development of techniques for the synthesis and functionalization of bis(indolyl)methanes have attracted more and more attention in recent years. Objective: To study the synthesis and biological activity of heterocyclic substituted bis(indolyl)methanes. Materials and Methods: A series of heterocyclic substituted bis(indolyl)methanes (3a-3p) have been prepared by condensation reaction of indole and heterocyclic aldehydes catalyzed by boron trifluoride etherate with high yields. Preliminary in vitro anti-inflammatory in lipopolysaccharide (LIPS)-stimulated RAW-264.7 macrophages and cytotoxic activity against human tumor cell lines (A549, Hela and SGC7901) by MTT assay were tested. Results: The result indicated that heterocyclic substituted bis(indolyl)methanes showed good anti-inflammatory and selective cytotoxic activity. Especially, compounds 3o, 3p and 3q displayed similar inhibitory effect on the generation of NO to positive control dexamethasone, and compound 3q displayed similar selective cytotoxic activity to 5-FU. Conclusion: Heterocyclic substituted bis(indolyl)methanes may be used as potential anti-inflammatory and anticancer leads.

Three new mononuclear complexes of the type [M(L)(2)](ClO4)(2), with Ni(II) (1), Cu(II) (2) and Zn(II) (3) ions, have been synthesized and structurally characterized, using the N,N,O donor Schiff base ligand 2-[(phenyl-pyridine-2-yl-methylene)-amino]-ethanol]. An X-ray diffraction study reveals identical crystallization patterns for each of the complexes, in the monoclinic crystal system with the C2/c space group. Interestingly, the synthesized complexes are isostructural in nature. The complexes showed biomimetic phenoxazinone synthase-like activity for the aerial oxidation of o-aminophenol to phenoxazine-2-one, separately in methanol (MeOH) and acetonitrile (ACN) solutions. For each of the complexes, the turnover numbers were found to be higher in MeOH than in ACN. The highest catalytic activity between among the three complexes was found to be for complex 2. Expanding the chemical diversity of metal complexes provides a robust platform to generate functional bioactive reagents. To access an excellent repository of metal-based compounds for probe/drug discovery, we capitalized on the rich chemistry of gold to create organometallic gold(III) compounds by ligand tuning. We obtained novel organogold(III) compounds bearing a 1,2-bis(diphenylphosphino)benzene ligand, providing structural diversity with optimal physiological stability. Biological evaluation of the lead compound AuPhos-89 demonstrates mitochondrial complex I-mediated alteration of the mitochondrial electron transport chain (ETC) to drive respiration and diminish cellular energy in the form of adenosine triphosphate (ATP). Mechanism-of-action efforts, RNA-Seq, quantitative proteomics, and NCI-60 screening reveal a highly potent anticancer agent that modulates mitochondrial ETC. AuPhos-89 inhibits the tumor growth of metastatic triple negative breast cancer and represents a new strategy to study the modulation of mitochondrial respiration for the treatment of aggressive cancer and other disease states where mitochondria play a pivotal role in the pathobiology.

A chemical sensor composed of pyridoxal hydrochloride schiff base based on diarylethene (1O) was synthesized. Its photochemical properties and selectivity to ions were further studied. The chemosensor could detect cyanide effectively and is almost undisturbed by other ions. When titrating CN-, the reaction aroused a distinct change in the absorption spectrum with the color change from transparent to yellow, and the fluorescence intensity centered at 562 nm was increased 68 folds. It also exhibited a good fluorescence sensing of Mg(2+ )with high selectivity and sensitivity. Upon addition of Mg2+, its emission intensity enhanced 110 folds, with the color change from dark to bright blue. Its good spectral response could be applied to molecular logic circuit. Moreover, the chemosensor could be made into test paper strips for the qualitative and quantitative detection of CN- and Mg2+.

An expeditious, high-yielding synthesis of rare alpha-fluoroepoxides and alpha-fluoroaziridines through the addition of the unkown fluoroiodomethyllithium (LiCHIF)-formed via deprotonation the commercially available fluoroiodomethane with a lithium amide base-to carbonyl-like compounds is documented. The ring-closure reactions, leading to alpha-fluorinated three-membered heterocycles, rely on the diversely reactive C-I and C-F bonds. Excellent chemoselectivity was observed in the presence of highly sensitive functionalities-aldehyde, ketone, nitrile, alkene-which remained untouched during the homologation sequence.

Aberrant activation of ERK signaling pathway usually leads to oncogenesis, and small molecular agents targeting this pathway are impeded by the emergence of drug resistance due to reactivation of ERK signaling. Compound DEL-22379 has been reported to inhibit ERK dimerization which was unaffected by drug-resistant mechanism reactivating the ERK signaling. Here, we discussed a structure-activity relationship study of DEL-22379. Forty-seven analogues were designed and synthesized. Each synthesized compound was biologically evaluated for their inhibitory rates on several tumor cell lines and compounds with high inhibitory rates were further evaluated for IC50 values. The structure-activity relationship of idolin-2-one scaffold and the impact of Z/E configuration on potency were discussed. Potential safety of two synthesized analogues was investigated and in silico docking study of five compounds was performed to understand the structural basis of ERK dimerization inhibition. Although pyridine derivatives are versatile supporting ligands in catalysis, the development of their chiral versions has been relatively limited. Herein, we report the design, synthesis, and proof-of-concept application of a structurally tunable chiral pyridine framework featuring an annulated compact ring system. Using an N,B-bidentate ligand skeleton containing the chiral pyridine moiety, we have developed an enantioselective iridium-catalyzed desymmetrizing C-H borylation reaction of diaryl(2-pyridyl)methane compounds with up to 96% ee and 93% yield. The resulting borylation products could be readily transformed into various chiral tri(hetero)arylmethane compounds. Density functional theory investigations revealed that the two chair conformations of the flexible ketal motif both favored the enantiomer that was consistent with experimental results. This work has thus introduced an effective and tunable chiral pyridine ligand framework that may be used in many catalytic asymmetric transformations.

The kinetics and mechanism of condensation of pyridoxal hydrochloride with L-alpha-asparagine, L-alpha- and D-alpha-aspartic acids are analyzed via UV spectroscopy and polarimetry. It is found that L-alpha-asparagine containing alpha-NH2 and gamma-NH2 groups interacts with pyridoxal via the gamma-NH2 group, forming Schiff bases that are resistant to chemical transformations. Rearrangement produces Schiff bases that form the cyclic structure from the amino acid moiety. L-alpha- and D-alpha-aspartic acids interacting with pyridoxal via alpha-NH2 groups create Schiff bases that form quinoid structures after elimination of alpha-hydrogen or CO2. Their subsequent hydrolysis results in pyridoxamine, alpha-ketoacids, and aldehyde acids, respectively. Schemes of the condensation mechanisms of L-alpha-asparagine, L-alpha-, D-alpha-aspartic acids with pyridoxal hydrochloride are proposed.

Tertiary alpha-hydroxyphosphine oxides have been synthesized via the catalyst- and solvent-free reaction between available secondary phosphine oxides and aliphatic, aromatic and heteroaromatic ketones at 20-62 degrees C in near to 96-98% yield. An organocatalyzed aerobic benzylic C-H oxidation of alkyl and aryl heterocycles has been developed. This transition metal-free method is able to overcome the electron-withdrawing effect as well as product-inhibition effects in heterobenzylic radical oxidation.
