Rummelt, Stephan M.;Peterson, Paul O.;Zhong, Hongyu;Chirik, Paul J. research published 《 Oxidative Addition of Aryl and Alkyl Halides to a Reduced Iron Pincer Complex》, the research content is summarized as follows. The two-electron oxidative addition of aryl and alkyl halides to a reduced Fe dinitrogen complex with a strong-field tridentate pincer ligand was demonstrated. Addition of iodobenzene or bromobenzene to (3,5-Me2MesCNC)Fe(N2)2 (3,5-Me2MesCNC = 2,6-(2,4,6-Me-C6H2-imidazol-2-ylidene)2-3,5-Me2-pyridine) resulted in rapid oxidative addition and formation of the diamagnetic, octahedral Fe(II) products (3,5-Me2MesCNC)Fe(Ph)(N2)(X), where X = I or Br. Competition experiments established the relative rate of oxidative addition of aryl halides as I > Br > Cl. A linear free energy of relative reaction rates of electronically differentiated aryl bromides (ρ = 1.5) was consistent with a concerted-type pathway. The oxidative addition of alkyl halides such as Me-, isobutyl-, or neopentyl halides was also rapid at room temperature, but substrates with more accessible β-H positions (e.g., 1-bromobutane) underwent subsequent β-hydride elimination. Cyclization of an alkyl halide containing a radical clock and epimerization of neohexyl iodide-d2 upon oxidative addition to (3,5-Me2MesCNC)Fe(N2)2 are consistent with radical intermediates during C(sp3)-X bond cleavage. Importantly, while C(sp2)-X and C(sp3)-X oxidative addition produces net two-electron chem., the preferred pathway for obtaining the products is concerted and stepwise, resp.

Ruzi, Rehanguli;Liu, Kai;Zhu, Chengjian;Xie, Jin research published 《 Upgrading ketone synthesis direct from carboxylic acids and organohalides》, the research content is summarized as follows. A photoredox, nickel and phosphoranyl radical synergistic cross-electrophile coupling of com. available chems., aromatic acids RCO2H (R = 4-methylphenyl, thiophen-3-yl, naphthalen-2-yl, etc.) and aryl/alkyl bromides R1Br (R1 = 6-(trifluoromethyl)pyridin-3-yl, 2-cyanoethyl, quinolin-3-yl, etc.) was reported. This allows for concise synthesis of highly functionalized ketones RC(O)R1directly, without the preparation of activated carbonyl intermediates or organometallic compounds, and thus complements the conventional Weinreb ketone synthesis. Use of the appropriate photocatalyst, ligand amount and solvents can match the reaction rate required by any simple catalytic cycle. The practicality and synthetic robustness of the reaction are illustrated by the facile synthesis of complex ketones from readily available feedstock chems.

J.;Grayson, Matthew N.;Cresswell, Alexander J. research published 《 Photocatalytic α-Tertiary Amine Synthesis via C-H Alkylation of Unmasked Primary Amines》, the research content is summarized as follows. A practical, catalytic entry to α,α,α-trisubstituted (α-tertiary) primary amines by C-H functionalization has long been recognized as a critical gap in the synthetic toolbox. The authors report a simple and scalable solution to this problem that does not require any in situ protection of the amino group and proceeds with 100% atom-economy. The authors’ strategy, which uses an organic photocatalyst in combination with azide ion as a hydrogen atom transfer (HAT) catalyst, provides a direct synthesis of α-tertiary amines, or their corresponding γ-lactams. The authors anticipate that this methodol. will inspire new retrosynthetic disconnections for substituted amine derivatives in organic synthesis, and particularly for challenging α-tertiary primary amines.

Nazari Montazer, Mohammad;Asadi, Mehdi;Bahadorikhalili, Saeed;Hosseini, Faezeh Sadat;Amanlou, Arash;Biglar, Mahmood;Amanlou, Massoud research published 《 Design, synthesis, docking study and urease inhibitory activity evaluation of novel 2-((5-amino-1,3,4-thiadiazol-2-yl)thio)-N-arylacetamide derivatives》, the research content is summarized as follows. Novel 2-((5-amino-1,3,4-thiadiazol-2-yl)thio)-N-arylacetamide derivatives I [R = 4-MeOC6H4, 4-methylisoxazol-3-yl, 5-chloro-2-pyridyl, etc.] were designed, synthesized and evaluated in vitro for their urease inhibitor activities. The compounds were synthesized efficiently in three steps in high isolated yields from amines, 2-chloroacetyl chloride, hydrazinecarbothioamide and carbon disulfide. The mol. docking simulation were performed using AutoDock4 by docking all synthesized compound and standard inhibitors into the crystal structure of Jack bean urease. Comparison between the urease inhibitory activity of compounds with the IC50 of (2.85-5.83μM) and thiourea and hydroxyurea as standards inhibitors with the IC50 of (22.00 and 100.00μM, resp.) proved the high activity of the synthesized compounds against the mentioned enzyme. Docking results were in good agreement with exptl. results and indicate that synthesized compounds could interact well with the active site of the urease enzyme and among all; compound I [R = 5-methyl-2-pyridyl] showed more favorable interactions with the active site which confirm its great inhibitory activity with IC50 of 2.85μM. Therefore, compound I [R = 5-methyl-2-pyridyl] might be a promising candidate for further evaluation.

Computed Properties of 766-11-0, 5-Bromo-2-fluoropyridine is a useful research compound. Its molecular formula is C5H3BrFN and its molecular weight is 175.99 g/mol. The purity is usually 95%.
5-Bromo-2-fluoropyridine is a boronic acid that has been shown to react with iodides and form 5-bromo-2-fluoroiodobenzene. The reaction of 5-bromo-2-fluoropyridine with benzene gives the same product as the reaction with 1,3,5-trioxane. The UV absorption of 5-bromo-2-fluoropyridine is found at 230 nm and 260 nm. It also has an absorption band in the infrared region at 1625 cm−1. Vibrational progressions have been observed for this molecule, which are due to dipole moments and electron density distributions in the molecule.

Neumann, Theresa;Benajiba, Lina;Goering, Stefan;Stegmaier, Kimberly;Schmidt, Boris research published 《 Evaluation of Improved Glycogen Synthase Kinase-3α Inhibitors in Models of Acute Myeloid Leukemia》, the research content is summarized as follows. The challenge for glycogen synthase kinase-3 (GSK-3) inhibitor design lies in achieving high selectivity for one isoform over the other. The therapy of certain diseases, such as acute myeloid leukemia (AML), may require α-isoform specific targeting. The scorpion shaped GSK-3 inhibitors developed by the group achieved the highest GSK-3α selectivity reported so far but suffered from insufficient aqueous solubility This work presents the solubility-driven optimization of the isoform-selective inhibitors using a scorpion shaped lead. Among 15 novel compounds, compound I showed high activity against GSK-3α/β with the highest GSK-3α selectivity reported to date. Compound I was profiled for bioavailability and toxicity in a zebrafish embryo phenotype assay. Selective GSK-3α targeting in AML cell lines was achieved with compound I, resulting in a strong differentiation phenotype and colony formation impairment, confirming the potential of GSK-3α inhibition in AML therapy.
