A new series (6) of C-5-curcuminoid derivatives (2E,6E-2,6-dibenzylidene-4-hydroxycyclohexanones) is described here with their evaluation for in vitro antiproliferative activities. Evaluation of 31 compounds against human A2780 (ovarian), C33A (cervix) and MDA-MB-231 (breast) cancer cell lines was performed to obtain structure activity relation data. The best performer was (2E,6E)-2,6-bis(3 ‘-nitrobenzylidene)-4-hydroxycyclohexanone (6h) with IC50 values of 0.68 mu M (A2780), 0.69 mu M (C33A) and 0.92 mu M (MDA-MB-231) compared to cisplatin with 1.30 mu M, 3.69 mu M and 19.13 mu M, respectively. According to calculated physicochemical properties some members in series 6, namely (2E,6E)-2,6-bis[(4 ‘-pyridinyl)methylene]-4-hydroxycyclohexanone (6p) [IC50 = 0.76 mu M (A2780), 2.69 mu M (C33A), 1.28 mu M (MDA-MB-231)] seem to have improved bioavailability compared to curcumin. Selected members of series 6 were involved in circular dichroism spectroscopic measurements in order to determine their interaction with natural DNA. Based on these data, we conclude that these derivatives do not bind to DNA in vitro. A proposal is summarized based on mass spectrometric assessment for fingerprint analysis in biological research of such C-5-curcuminoids.

Purpose: We investigated Nw-hydroxy L-Arginine (NOHA) predictive response in serous ovarian carcinoma based on estrogen-hormone receptor expression status; and assessed the distinctive NOHA response between estrogen-receptor-negative (ER-) tumor subtypes of ovarian and breast cancer. Materials/methods: Three-dimensional (3D) spheroids models of ER- and estrogen-receptor-positive (ER+) from breast and ovarian tumor, cultured for 9 weeks, were assayed for cellular levels of inducible nitric oxide synthase (NOS2), nitric oxide (as total nitrite) and L-Arginine, and compared to NOHA in culture medium. Statistical difference was set at p < 0.01. Results: Nine-week in vitro studies showed a progressive NOHA reduction in culture medium by at least 0.4-0.8 fold, and 0.65-0.92 fold only in the ER-breast tumor and ER-ovarian tumor 3D spheroids, respectively; with increases in cellular NOS2 and nitric-oxide levels, by at least 1.0-2.45 fold in both ER-tumor subtype 3D spheroids (p < 0.01; n = 6). Within ER-subtypes, medium NOHA decreased by >= 38.9% in ovarian cancer over breast cancer 3D-spheroids, with cellular increases in NOS2 (by >= 17.4%), and nitric oxide (by >= 18.8%). Cellular L-Arginine to medium NOHA ratio was higher, and by at least 6.5-22.5 fold in ER-breast tumor 3D-spheroids, and at least 10-70 fold in ER-ovarian tumor 3D spheroids, than in ER+ and control conditions; and was >= 48% higher in ER-ovarian cancer than in ER-breast cancer 3D-spheroids. Conclusions: The present study shows NOHA as a sensitive and selective indicator differentiating and distinguishing ER-subtypes based on the tumor grade.

A three-descriptor quantitative structure-activity/toxicity relationship (QSAR/QSTR) model was developed for the skin permeability of a sufficiently large data set consisting of 274 compounds, by applying support vector machine (SVM) together with genetic algorithm. The optimal SVM model possesses the coefficient of determination R-2 of 0.946 and root mean square (rms) error of 0.253 for the training set of 139 compounds; and a R-2 of 0.872 and rms of 0.302 for the test set of 135 compounds. Compared with other models reported in the literature, our SVM model shows better statistical performance in a model that deals with more samples in the test set. Therefore, applying a SVM algorithm to develop a nonlinear QSAR model for skin permeability was achieved.

Structure-activity relationships for a series of small-molecule thiophenes resulted in potent and selective antagonism of human Complement C3a receptor. A series of bidendate pyridine-functionalized metal N-heterocyclic carbene (NHC) complexes with various wingtip substituents (R = methyl, phenyl and tert-butyl) had been prepared from silver oxide-mediated transmetalation method. The basic backbone of the NHC ligands were prepared in two step-wise reactions: reduction of the commercially available 2-benzoylpyridine, followed by halogenation to yield 2-(chloro(phenyl) methyl) pyridine. Subsequent reactions with different imidazoles gave the desired imidazolium salts (or NHC precursors) with different R groups at the N wingtip (R = Me, Ph, t-Bu). Transmetalation of the imidazolium salts into platinum NHC complexes were achieved by using Ag2O mediated method with Pt(cod)Cl-2 as the transmetalating agent. On the other hand, nickel NHC complexes were obtained via one pot synthesis method with nickel chloride as the transmetalating agent and reflux in the presence of a base. Following recrystallisation, the molecular structure of Pt NHC complex (+/-)-5a had been successfully elucidated via single crystal X ray diffraction (XRD) analysis. The six-membered ring of the platinum NHC complexes adopted a boat conformation with the phenyl ring arranged at the axial position. Meanwhile, two unexpected nickel complexes, i.e. a simple nickel coordination complex and a nickelate complex, were obtained and studied using XRD. These Pt NHC complexes and unexpected Ni complexes were evaluated for their in vitro antimicrobial and anticancer activities against a panel of pathogenic microorganisms and three selected human cancer cell lines, including breast (MCF7), colon (HCT116) and oral (H103) cancer cells. Generally, the Pt NHC complexes displayed enhanced antimicrobial activities upon coordination to metals, as compared to the corresponding imidazolium salts i.e. NHC precursors, with minimum inhibitory concentration (MIC) values in micromolar (mM) range. A couple of platinum NHC complexes synthesized exhibited antimicrobial activities with MIC as low as 2 mM, which are comparable to silver NHC complexes with renowned antimicrobial profiles. Similarly, upon coordination to the metals, the cytotoxic effects of the platinum NHC complexes increased significantly as compared to the imidazolium salts. Among all, platinum NHC complex (+/-)-5c displayed significant cytotoxicities towards the cancer cells with IC50 values that are two to three times lower than that of the anticancer drug cisplatin. Cobalt complexes containing a salen-type pyridoxal ligand with pyridine were synthesized as a new Co (III) catalytic system for the cycloaddition of carbon dioxide. Two cobalt(III) complexes possessing a salen-type pyridoxyl ligand were synthesized by the reaction of pyridoxal ligands (pyr(2)en = (N,N’-bis (pyridoxylideneiminato)ethylene) and pyr(2)cy = (N,N’-bis(pyridoxylideneiminato)cyclohexane)) and Co (OAc)(2) and characterized by various analytical methods, including infrared spectroscopy and high-resolution mass analysis. Single-crystal X-ray crystallography analysis confirmed that the cobalt pyr(2)en complex had a distorted octahedral structure: the tetradentate Schiff base ligand binds the cobalt metal in one plane, and the metal center adopts an octahedral geometry by the additional coordination of acetate and dimethyl sulfoxide. The synthesized complexes were used as catalysts in the cycloaddition of carbon dioxide (CO2) to propylene oxide. Antibiotic resistance remains a major global public health threat that requires sustained discovery of novel antibacterial agents with unexploited scaffolds. Structure-activity relationship of the first-generation aryl isonitrile compounds we synthesized led to an initial lead molecule that informed the synthesis of a second-generation of aryl isonitriles. From this new series of 20 compounds, three analogues inhibited growth of methicillin-resistant Staphylococcus aureus (MRSA) (from 1 to 4 mu M) and were safe to human keratinocytes. Compound 19, with an additional isonitrile group exhibited improved activity against MRSA compared to the first-generation lead compound. This compound emerged as a candidate worthy of further investigation and further reinforced the importance of the isonitrile functionality in the compounds’ anti-MRSA activity. In a murine skin wound model, 19 significantly reduced the burden of MRSA, similar to the antibiotic fusidic acid. In summary, 19 was identified as a new lead aryl isonitrile compound effective against MRSA.

N-Unprotected ketimines are useful substrates and intermediates for synthesizing valuable nitrogen-containing compounds, but their potential applicability is limited by the available synthetic methods. To address this issue, we report a scandium(III) triflate catalyzed direct synthesis of N-unprotected ketimines. Using commercially available reagents and Lewis acid catalysts, ketones were directly transformed into the corresponding N-unprotected ketimines in high yields with broad functional group tolerance, even in multigram scales. The reactions were readily applicable for one-pot synthesis of important compounds such as a glycine Schiff base without isolation of N-unprotected ketimine intermediates. We report a mild palladium-catalyzed method for the selective allylation of 4-alkylpyridines in which highly basic pyridylic anions behave as soft nucleophiles. This method exploits alkylidene dihydropyridines, which are semi-stable intermediates readily formed using a ‘soft-enolization’ approach, in a new mechanistic manifold for decarboxylative allylation. Notably, the catalytic generation of pyridylic anions results in a substantially broader functional group tolerance compared to other pyridine allylation methods. Experimental and theoretical mechanistic studies strongly suggest that pyridylic anions are indeed the active nucleophiles in these reactions, and that they participate in an outer-sphere reductive elimination step.
