Directed evolution of enzymes for the asymmetric reduction of prochiral ketones to produce enantio-pure secondary alcohols is particularly attractive in organic synthesis. Loops located at the active pocket of enzymes often participate in conformational changes required to fine-tune residues for substrate binding and catalysis. It is therefore of great interest to control the substrate specificity and stereochemistry of enzymatic reactions by manipulating the conformational dynamics. Herein, a secondary alcohol dehydrogenase was chosen to enantioselectively catalyze the transformation of difficult-to-reduce bulky ketones, which are not accepted by the wildtype enzyme. Guided by previous work and particularly by structural analysis and molecular dynamics (MD) simulations, two key residues alanine 85 (A85) and isoleucine 86 (I86) situated at the binding pocket were thought to increase the fluctuation of a loop region, thereby yielding a larger volume of the binding pocket to accommodate bulky substrates. Subsequently, site-directed saturation mutagenesis was performed at the two sites. The best mutant, where residue alanine 85 was mutated to glycine and isoleucine 86 to leucine (A85G/I86L), can efficiently reduce bulky ketones to the corresponding pharmaceutically interesting alcohols with high enantioselectivities (similar to 99% ee). Taken together, this study demonstrates that introducing appropriate mutations at key residues can induce a higher flexibility of the active site loop, resulting in the improvement of substrate specificity and enantioselectivity.

A novel fluorescent probe, TCF-IPY, has been designed and developed for highly selective and ratiometric detection of GSH based on the extend of the pi-conjugation system of imidazo[1,5-alpha]pyridine IPY-CHO. Upon addition of GSH, TCF-IPY displayed remarkable fluorescence variations (approximate 949-fold ratio changes) from 603 nm to 475 nm, thereby enabling GSH detection in a fine ratiometric manner. Moreover, TCF-IPY exhibited excellent selectivity toward GSH over other biologically related species including Cys and Hcy. The huge blue shifts both in absorption (259 nm) and emission spectra (128 nm), low cytotoxicity, high sensitivity (97 nM) and rapid response (240 s) of TCF-IPY make it a robust molecular tool for endogenous GSH investigation. Furthermore, TCF-IPY was also successfully used for ratiometric imaging of endogenous GSH in living MCF-7 cells and zebrafish.

In this study, the chemical and sensory profiles of 42 different nonalcoholic beer (NAB) brands/styles already on the global market and produced through several different brewing techniques were evaluated. A trained panel (i.e., 11 panelists) performed standard-driven descriptive and check-all-that-apply analyses in triplicate to sensorially characterize the aroma and taste/mouthfeel profiles of 42 commercial NABs, a commercial soda, and a commercial seltzer water (n = 44). These beers were also chemically deconstructed using several different analytical techniques targeting volatile and nonvolatile compounds. Consumer analysis (n = 129) was then performed to evaluate the Northern Californian consumer hedonic liking of a selection (n = 12) of these NAB brands. These results provide direction to brewers and/or beverage producers on which techniques they should explore to develop desirable NAB offerings and suggest chemical targets that are indicators of specific flavor qualities and/or preference for American consumers.

Oxidized products have become increasingly valuable as building blocks for a wide variety of different processes and fine chemistry, especially in the benzylic position. We report herein a sustainable protocol for this transformation through C-H functionalization and is performed using electrochemistry as the main power source and tert-butyl hydroperoxide as the radical source for the C-H abstraction. The temperature conditions reported here do not increase above 50 degrees C and use an aqueous-based medium. A broad substrate scope is explored, along with bioactive molecules, to give comparable and increased product yields when compared to prior reported literature without the use of electrochemistry.

Three hydroxymethyl bridged Cu(ii) complexes of a pyridoxal Schiff base ligand 4-((E)-(2-(pyridin-2-yl)ethylimino)methyl)-5-(hydroxymethyl)-2-methylpyridin-3-ol (LH) have been synthesized and characterized on the basis of spectroscopic, elctrochemical and structural properties. The X-ray crystal structures of the complexes reveal dual denticity of the ligand, bidenticity in the absence of a co-ligand as in complex1, and tridenticity in the presence of a co-ligand such as SCN-/N(CN)(2)(-)as in complexes2and3. The complexes, though binuclear in the solid state, exist as a monomeric unit in solution due to the exceptionally long axial Cu-O-hydroxymethyl(2.4-2.5 angstrom) bond. All three complexes show efficient catalytic activities towards the aerial oxidation of 3,5-ditertiarybutylcatechol (DTBCH2) withk(cat)values of 5.38 x 10(4)h(-1), 1.18 x 10(5)h(-1)and 1.06 x 10(5)h(-1)in methanol. Complexes1and2also act as a selective sulphide ion sensor withK(b)values of 6.6 x 10(3)M(-1)and 8.1 x 10(3)M(-1), respectively, while their respective L.O.D. values are 3.4 mu M and 3.2 mu M.

Two magnetic nano catalysts of nickel and copper, Fe3O4@SiO2@DOP-BenPyr-M(II), (M=Ni and Cu) have been synthesized. These catalysts were applied as recoverable, efficient and new heterogeneous catalysts for the high yielding and room temperature one-pot procedure of selective oxidation of sulfides to sulfoxides and oxidative coupling of thiols to disulfides. In addition, the catalytic activity of Fe3O4@SiO2@DOP-BenPyr-Ni(II) was investigated as heterogeneous nanocatalyst for synthesis of 2,3-dihydroquinazolin-4(1H)-ones, 5-substituted 1H-tetrazoles and polyhydroquinolines. The synthesized catalysts were characterized by FT-IR, TGA, XRD, VSM, EDX, ICP and SEM techniques. These catalysts were recovered by an external magnet and reused several times without significant loss of catalytic efficiency. [GRAPHICS] .

Herein, we demonstrate an environmental-friendly and safe electrocatalytic strategy for the selective oxidation of benzyl group to ketones at ambient conditions, while using water as the sole oxygen source. Water addition reduces the onset potential of anodic C-H oxidation, and produces 1-tetralone with satisfying conversion and excellent ketone to alcohol ratio. Layered MnO2 catalysts (with rich oxygen vacancies) further adjust the water affinity and facilitate the oxidation, leading to a significantly improved faradaic efficiency.
