A new coumarin based fluorescent switch (HCBP) has been fabricated which displays high selective sensing towards Al3+ among other metal cations at physiological pH. On gradual addition of Al3+ specifically, HCBP shows a brilliant turn-on emission enhancement of about similar to 13-fold with about 50 nm red shift at 481 nm in MeOH/H2O (1/1, v/v) solution. Herein, we realized the simultaneous online detection of six tobacco alkaloids (TM) by in-tube solid-phase microextraction (In-tube SPME) coupled with mass spectrometry by a rapid, sensitive, and matrix effect-free method requiring no chromatographic separation and only minimal sample pre-treatment. A poly (N-isopropylacrylamide-co-divinylbenzene-co-N, N’-methylenediacrylamide) [Poly (NIPAAm-co-DVB-co-MBAA)] monolithic column was designed according to the chemical structures of selected TM and used as an extraction medium engaging in hydrophobic, pi-pi, and hydrogen bonding interactions with analytes, allowing them to be effectively extracted. A number of important parameters were systematically optimized to achieve maximal extraction efficiency. The ion intensity of the TM signals obtained by in-tube SPME-MS were higher than the direct MS mode by about 400 folds with the signal-to-noise ratio improved by 2-7 folds. The detection limits of the six TM were determined as 1.99-4.06 ng g(-1), with good linearity with correlation coefficients exceeding 0.99 obtained under optimal extraction conditions. Besides, TA recoveries in cigarette tobacco spiked at three concentration levels were in the range of 76.4-100.2%, and the corresponding RSDs (n = 5) were obtained as 4.32-7.16%. The extraction performance of the poly (NIPAAm-co-DVB-co-MBAA) monolithic column was well reproducible, with intra- or inter-day precision RSDs determined not to exceed 7.38%. Finally, no marked matrix effects were observed when the developed method was applied to the analysis of both high-abundance and trace level TM in practical samples, and the above technique was therefore concluded to be well suited for the detection of TAs in cigarette tobacco or other products.

We herein report the fabrication of a bifunctional iron nanocomposite catalyst, in which two catalytically active sites of Fe-N-x and Fe phosphate, as oxidation and Lewis acid sites, were simultaneously integrated into a hierarchical N,P-dual doped porous carbon. As a bifunctional catalyst, it exhibited high efficiency for direct oxidative cleavage of alkenes into ketones or their oxidation into 1,2-diketones with a broad substrate scope and high functional group tolerance using TBHP as the oxidant in water under mild reaction conditions. Furthermore, it could be easily recovered for successive recycling without appreciable loss of activity. Mechanistic studies disclose that the direct oxidation of alkenes proceeds via the formation of an epoxide as intermediate followed by either acid-catalyzed Meinwald rearrangement to give ketones with one carbon shorter or nucleophilic ring-opening to generate 1,2-diketones in a cascade manner. This study not only opens up a fancy pathway in the rational design of Fe-N-C catalysts but also offers a simple and efficient method for accessing industrially important ketones and 1,2-diketones from alkenes in a cost-effective and environmentally benign fashion.

Despite huge progress in hormonal therapy and improved in vitro fertilization methods, the success rates in infertility treatment are still limited. A recently discovered mechanism revealed the interplay between the plasma protein fetuin-B and the cortical granule-based proteinase ovastacin to be a novel key mechanism in the regulation of fertilization. Upon sperm-egg fusion, cleavage of a distinct zona pellucida component by ovastacin destroys the sperm receptor, enhances zona robustness, and eventually provides a definitive block against polyspermy. An untimely onset of this zona hardening prior to fertilization would consequently result in infertility. Physiologically, this process is controlled by fetuin-B, an endogenous ovastacin inhibitor. Here we aimed to discover small-molecule inhibitors of ovastacin that could mimic the effect of fetuin-B. These compounds could be useful lead structures for the development of specific ovastacin inhibitors that can be used in infertility treatment or in vino fertilization.

Curcumin and trans-cinnamaldehyde are acrolein-based Michael acceptor compounds that are commonly found in domestic condiments, and known to cause cancer cell death via redox mechanisms. Based on the structural features of these compounds we designed and synthesized several 2-cinnamamido-N-substituted-cinnamamide (bis-cinnamamide) compounds. One of the derivatives, (Z)-2-[(E)-cinnamamido]-3-phenyl-N-propylacrylamide 8 showed a moderate antiproliferative potency (HCT-116 cell line inhibition of 32.0 mu M), no inhibition of normal cell lines C-166, and proven cellular activities leading to apoptosis. SAR studies led to more than 10-fold increase in activity. Our most promising compound, [(Z)-3-(1H-indol-3-yl)-N-propyl-2-[(E)-3-(thien-2-yl)propenamido) propenamide] 45 killed colon cancer cells at IC50 = 0.89 mu M (Caco-2), 2.85 mu M (HCT-116) and 1.65 mu M (HT -29), while exhibiting much weaker potency on C-166 and BHK normal cell lines (IC50 = 71 mu M and 77.6 mu M, respectively). Cellular studies towards identifying the compounds mechanism of cytotoxic activities revealed that apoptotic induction occurs in part as a result of oxidative stress. Importantly, the compounds showed inhibition of cancer stem cells that are critical for maintaining the potential for self-renewal and stemness. The results presented here show discovery of covalently acting Michael addition compounds that potently kill cancer cells by a defined mechanism, with prominent selectivity profile over non-cancerous cell lines.

For the first time, the [4 + 3] or [2 + 1] annulation of crotonate-derived sulfur ylides with arylidenemalononitrile or arylidene-1H-indene-1,3(2H)-dione is reported using Na(2)CO(3)as the base. Small-scale culture of animal cells in suspension is of importance for many applications. At a small-scale, fed-batch is achieved either by manual bolus feeding or the use of liquid handling robots. In this study, we report an alternate application of a hydrogel for in situ continuous delivery of a nutrient feed comprising 18 amino acids, vitamins, antioxidants, and trace elements. We show that amino acid release is sustained for at least seven days. Importantly, release rates of individual amino acids can be independently modulated by changing their loading. We demonstrate the application of this hydrogel for complete in situ feeding of nutrients to a suspension adapted CHO cell line expressing IgG leading to 2.7-fold and 4-fold improvement in integral viable cell density (IVCD) and volumetric productivity respectively. This is similar to improvements obtained by bolus liquid feeding. Further, supplying glucose from the same hydrogel to eliminate manual feeding led to a 1.8-fold increase in IVCD accompanied by a 3-fold increase in volumetric productivity as compared to batch culture. In summary, this study provides a proof-of-concept that hydrogels can enable completely closed in situ feeding for mammalian cell culture requiring no external intervention. Such continuous in situ delivery can potentially enable closed culture systems maintaining nutrients at low levels mimicking physiological concentrations.

The interaction between pyridoxal hydrochloride (HQ) and apoCopC was investigated using Fourier transform infrared spectroscopy (FTIR), isothermal titration calorimetry (ITC), circular dichroism (CD), fluorescence spectroscopy, three-dimensional (3D) fluorescence spectroscopy, fluorescence lifetime, TNS fluorescence and docking methods. FTIR, CD, TNS fluorescence and fluorescence lifetime experiments suggested that the apoCopC conformation was altered by HQ with an increase in the random coil content and a reduction in the beta-sheet content. In addition, the data from fluorescence spectroscopy, 3D fluorescence spectroscopy and molecular docking revealed that the binding site of HQ was located in the hydrophobic area of apoCopC, and a redshift of the HQ fluorescence spectra was observed. Furthermore, ITC and fluorescence quenching data manifested that the binding ratio of HQ and apoCopC was 1:1, and the forming constant was calculated to be (7.06 +/- 0.21) x 10(5) M-1. The thermodynamic parameters Delta H and Delta S suggested that the formation of a CopC-HQcomplex depended on the hydrophobic force. Furthermore, the average binding distance between tryptophan in apoCopC and HQ was determined by means of Forster non-radioactive resonance energy transfer and molecular docking. The results agreed well with each other. As a redox switch in the modulation of copper, the interaction of apoCopC with small molecules will affect the action of the redox switch. An efficient, reusable and stable binaphthyl stabilized Pd-nanoparticles (Pd-BNP) catalyzed the direct ortho-C-H hydroxylation of acetophenone oxime ethers under neutral and phosphine ligand-free condition has been developed. A readily available, economic, safe and greener oxidant oxone has been used in this direct ortho-hydroxylation. The scope of the reaction has been studied with various acetophenone oxime ethers including electron rich to electron deficient system and the reaction afforded only mono hydroxylated products in a highly regioselective manner. Several control experiment results confirmed that the oxone is the hydroxyl source. The Pd-BNP catalyst has been reused up to five times. The heterogeneous test confirmed that the reaction is catalyzed by the heterogeneous Pd-BNP catalyst.
