In this study Guar gum based Cu(II) Schiff’s base complex (GG-Cu) has been synthesized and characterized by FTIR, PXRD, UV-vis, TGA, XPS, FESEM, TEM, EDAX, solid-state NMR, Elemental mapping, CHNS and AAS analysis. This moiety has been found to be an efficient heterogeneous catalyst for selective oxidation reactions. Fifteen model reactions have been carried to establish the catalytic behavior of GG-Cu, and five of these yield novel products. The ease of separation of catalyst from the reaction mixture simply by filtration is an added advantage; furthermore the catalyst can be reused up to five times without significant loss of catalytic activity. The overall concept of developing newer, efficient and environmental benign catalysts with ease of separation and recycling ability has been successfully demonstrated. All of the isolated products were fully characterized on the basis of their physical and spectral data.

Thiourea-organocatalyzed Michael additions of diethyl malonate to various heteroaromatic nitroolefins (13 examples) have been studied under high-pressure (up to 800 MPa) and ambient pressure conditions. Triazole-based ligands, tris (triazolyl)methanol (Htbtm), bis (triazolyl)-phenylmethanol (Hbtm), and phenyl (pyridin-2-yl)(triazolyl)methanol (Hpytm), with differences in ligand denticity (i.e., bidentate and tridentate) and type of N donors (i.e., triazole and pyridine) were functionalized onto a silica support to produce the corresponding SiO2-L (L = tbtm, btm, pytm). Subsequent reactions with Pd (CH3COO)(2) in CH2Cl2 yielded Pd/SiO2-L. ICP-MS reveals that Pd loadings are higher with increased N loadings, resulting in the following trend: Pd/SiO2-tbtm (0.83 mmol Pd g(-1)) > Pd/SiO2-btm (0.65 mmol Pd g(-1)) ~ Pd/SiO2-pytm (0.63 mmol Pd g(-1)). Meanwhile, TEM images of the used Pd/SiO2-L catalysts after the first catalytic cycle show that the mean size of Pd NPs is highest with Pd/SiO2-pytm (8.5 +/- 1.5 nm), followed by Pd/SiO2-tbtm (6.4 +/- 1.6 nm) and Pd/SiO2-btm (4.8 +/- 1.3 nm). Based on TONs, catalytic studies toward aerobic oxidation of benzyl alcohol to benzaldehyde at 60 degrees C in EtOH showed that Pd/SiO2-pytm possessed the most active surface Pd(0) atoms, most likely as a result of more labile properties of the pyridine-triazole ligand compared to tris- and bis (triazolyl) analogs. ICP-MS and TEM analysis of Pd/SiO2-btm indicate minimal Pd leaching and similar average Pd NPs sizes after 1(st) and 5(th) catalytic runs, respectively, confirming that SiO2-btm is an efficient Pd NPs stabilizer. The Pd/SiO2-btm catalyst was also active toward aerobic oxidation of various benzyl alcohol derivatives in EtOH and could be reused for at least 7 reaction cycles without a significant activity loss.

General and efficient approaches for the synthesis of new 5-amino and 5-iminoimidazo[1,2-a] imidazoles were developed through a three-component reaction of 1-unsubstituted 2-aminoimidazoles with various aldehydes and isocyanides mediated by zirconium(IV) chloride. A series of novel glycosyl nitrogen-containing heterocycles derivatives were designed and synthesized. With the help of microwave, the reaction was carried out in water rapidly and afforded the target compounds in good yields. The acid catalysts that were essential for this kind of reaction under traditional heating method were avoided through this strategy. Preliminary biological evaluation showed that most of the compounds could inhibit the growth of A549 cells, but the inhibition of HepG-2 cells was relatively poor. Notably, compound 2i displayed the best potency with an IC50 value of 3.42 mu M against A549 cell lines, which is comparable with the common anticarcinogen paclitaxel (5.12 mu M). Molecular modeling studies suggested that 2i may bind to the ATP-binding site of epidermal growth factor receptor (EGFR), indicating a rational design strategy. These results provide a starting point for designing glycosyl nitrogen-containing heterocycles as the potential drugs in lung cancer therapy.

In vitro viability assays against a representative panel of human cancer cell lines revealed that polyamines L1a and L5a displayed remarkable activity with IC50 values in the micromolar range. Preliminary research indicated that both compounds promoted G1 cell cycle arrest followed by cellular senescence and apoptosis. The induction of apoptotic cell death involved loss of mitochondrial outer membrane permeability and activation of caspases 3/7. Interestingly, L1a and L5a failed to activate cellular DNA damage response. The high intracellular zinc-chelating capacity of both compounds, deduced from the metal specific Zinquin assay and ZnL2+ stability constant values in solution, strongly supports their cytotoxicity. These data along with quantum mechanical studies have enabled to establish a precise structure-activity relationship. Moreover, L1a and L5a showed appropriate drug-likeness by in silico methods. Based on these promising results, L1a and L5a should be considered a new class of zinc-chelating anticancer agents that deserves further development.

Cobalt complexes containing a salen-type pyridoxal ligand with pyridine were synthesized as a new Co (III) catalytic system for the cycloaddition of carbon dioxide. Two cobalt(III) complexes possessing a salen-type pyridoxyl ligand were synthesized by the reaction of pyridoxal ligands (pyr(2)en = (N,N’-bis (pyridoxylideneiminato)ethylene) and pyr(2)cy = (N,N’-bis(pyridoxylideneiminato)cyclohexane)) and Co (OAc)(2) and characterized by various analytical methods, including infrared spectroscopy and high-resolution mass analysis. Single-crystal X-ray crystallography analysis confirmed that the cobalt pyr(2)en complex had a distorted octahedral structure: the tetradentate Schiff base ligand binds the cobalt metal in one plane, and the metal center adopts an octahedral geometry by the additional coordination of acetate and dimethyl sulfoxide. The synthesized complexes were used as catalysts in the cycloaddition of carbon dioxide (CO2) to propylene oxide. Water-soluble vitamins like B3 (nicotinamide), B6 (pyridoxine), and B9 (folic acid) are of utmost importance in human health and disease, as they are involved in numerous critical metabolic reactions. Not surprisingly, deficiencies of these vitamins have been linked to various disease states. Unfortunately, not much is known about the physiological levels of B6 vitamers and vitamin B3 in an ethnically isolated group (such as an Emirati population), as well as their relationship with obesity. The aim of the present study was to quantify various B6 vitamers, as well as B3, in the plasma of obese and healthy Emirati populations and to examine their correlation with obesity. A sensitive and robust HPLC-MS/MS-based method was developed for the simultaneous quantitation of five physiologically relevant forms of vitamin B6, namely pyridoxal, pyridoxine, pyridoxamine, pyridoxamine phosphate, and pyridoxal phosphate, as well as nicotinamide, in human plasma. This method was used to quantify the concentrations of these vitamers in the plasma of 57 healthy and 57 obese Emirati volunteers. Our analysis showed that the plasma concentrations of nicotinamide, pyridoxal, and pyridoxamine phosphate in the obese Emirati population were significantly higher than those in healthy volunteers (p< 0.0001,p= 0.0006, andp= 0.002, respectively). No significant differences were observed for the plasma concentrations of pyridoxine and pyridoxal phosphate. Furthermore, the concentrations of some of these vitamers in healthy Emirati volunteers were significantly different than those published in the literature for Western populations, such as American and European volunteers. Materials/methods: Three-dimensional (3D) spheroids models of ER- and estrogen-receptor-positive (ER+) from breast and ovarian tumor, cultured for 9 weeks, were assayed for cellular levels of inducible nitric oxide synthase (NOS2), nitric oxide (as total nitrite) and L-Arginine, and compared to NOHA in culture medium. Statistical difference was set at p < 0.01. Results: Nine-week in vitro studies showed a progressive NOHA reduction in culture medium by at least 0.4-0.8 fold, and 0.65-0.92 fold only in the ER-breast tumor and ER-ovarian tumor 3D spheroids, respectively; with increases in cellular NOS2 and nitric-oxide levels, by at least 1.0-2.45 fold in both ER-tumor subtype 3D spheroids (p < 0.01; n = 6). Within ER-subtypes, medium NOHA decreased by >= 38.9% in ovarian cancer over breast cancer 3D-spheroids, with cellular increases in NOS2 (by >= 17.4%), and nitric oxide (by >= 18.8%). Cellular L-Arginine to medium NOHA ratio was higher, and by at least 6.5-22.5 fold in ER-breast tumor 3D-spheroids, and at least 10-70 fold in ER-ovarian tumor 3D spheroids, than in ER+ and control conditions; and was >= 48% higher in ER-ovarian cancer than in ER-breast cancer 3D-spheroids. Conclusions: The present study shows NOHA as a sensitive and selective indicator differentiating and distinguishing ER-subtypes based on the tumor grade.
