N-Unprotected ketimines are useful substrates and intermediates for synthesizing valuable nitrogen-containing compounds, but their potential applicability is limited by the available synthetic methods. To address this issue, we report a scandium(III) triflate catalyzed direct synthesis of N-unprotected ketimines. Using commercially available reagents and Lewis acid catalysts, ketones were directly transformed into the corresponding N-unprotected ketimines in high yields with broad functional group tolerance, even in multigram scales. The reactions were readily applicable for one-pot synthesis of important compounds such as a glycine Schiff base without isolation of N-unprotected ketimine intermediates. Preliminary mechanistic studies to clarify the reaction mechanism are also described.

Aryl and heteroaryl chiral carbinols are useful precursors in the synthesis of drugs. Lactobacillus paracasei BD87E6, which is obtained from a cereal based fermented beverage, was investigated as whole cell biocatalyst for the bioreduction of different ketones (including aromatic, hetero-aromatic and fused bicyclic ketone) into chiral carbinols, which can be used as a pharmaceutical intermediate. The study shows that bioreduction of aryl, heteroaryl and fused bicyclic ketone (1-5) to their corresponding chiral carbinols (1a-5a) in excellent enantioselectivity (>99%) with high yields. This study gave the first example for an enantiopure production of (S)-6-chlorochroman-4-ol (3a), which has many antioxidant activity, by a biological method. For asymmetric bioreduction of other prochiral ketones, these results open way to use of L. paracasei BD87E6 as biocatalysts. Also, the present process shows a hopeful and alternative green synthesis for the production of enantiopure carbinols in a mild, inexpensive and environmentally friendly process.

Three new mononuclear complexes of the type [M(L)(2)](ClO4)(2), with Ni(II) (1), Cu(II) (2) and Zn(II) (3) ions, have been synthesized and structurally characterized, using the N,N,O donor Schiff base ligand 2-[(phenyl-pyridine-2-yl-methylene)-amino]-ethanol]. An X-ray diffraction study reveals identical crystallization patterns for each of the complexes, in the monoclinic crystal system with the C2/c space group. Interestingly, the synthesized complexes are isostructural in nature. The complexes showed biomimetic phenoxazinone synthase-like activity for the aerial oxidation of o-aminophenol to phenoxazine-2-one, separately in methanol (MeOH) and acetonitrile (ACN) solutions. For each of the complexes, the turnover numbers were found to be higher in MeOH than in ACN. The highest catalytic activity between among the three complexes was found to be for complex 2. Three hydroxymethyl bridged Cu(ii) complexes of a pyridoxal Schiff base ligand 4-((E)-(2-(pyridin-2-yl)ethylimino)methyl)-5-(hydroxymethyl)-2-methylpyridin-3-ol (LH) have been synthesized and characterized on the basis of spectroscopic, elctrochemical and structural properties. The X-ray crystal structures of the complexes reveal dual denticity of the ligand, bidenticity in the absence of a co-ligand as in complex1, and tridenticity in the presence of a co-ligand such as SCN-/N(CN)(2)(-)as in complexes2and3. The complexes, though binuclear in the solid state, exist as a monomeric unit in solution due to the exceptionally long axial Cu-O-hydroxymethyl(2.4-2.5 angstrom) bond. All three complexes show efficient catalytic activities towards the aerial oxidation of 3,5-ditertiarybutylcatechol (DTBCH2) withk(cat)values of 5.38 x 10(4)h(-1), 1.18 x 10(5)h(-1)and 1.06 x 10(5)h(-1)in methanol. The chalcone and bis-chalcone derivatives have been synthesized under sonication conditions via Claisen-Schmidt condensation with KOH in ethanol at room temperature (20-89%). The structures were established on the basis of NMR, IR, Single-crystal XRD, and MS. The best compound 3u had inhibitory activity (IC50 = 7.50 mu M). The synthesis, the antioxidative properties, chemical reactivity descriptors supported in Density Functional Theory (DFT), acetylcholinesterase (AChE) inhibition and their potential binding modes, and affinity were predicted by molecular docking of a number of morpholine-chalcones and quinoline-chalcone. A series of bis-chalcones are also reported. Molecular docking and an enzyme kinetic study on compound 3u suggested that it simultaneously binds to the catalytic active site (CAS) and peripheral anionic site (PAS) of AChE. A new Schiff base receptor (2-amino-3-(((3-hydroxy-5-(hydroxymethyl)-2-methylpyridin-4-yl)methylene) amino)maleonitrile (GAL)) has been synthesized and such diaminomaleonitrile-based molecular framework is observed to be water soluble. GAL possesses both colorimetric and off-on fluorescent response in the presence of ClO-. The response time has been controlled within 6 min. The limit of detection (LOD) has been calculated to be 47.5 nM. The addition of Cu2+ can only induce clear color evolution from pale to deep yellow (LOD: 0.22 mu M) and no fluorescence changes are found. Moreover, its reliability and practicality are verified via the determination of ClO- in spiked samples of tap water and pond water. Schiff bases (SB) obtained from S-methyl dithiocarbazate and aromatic aldehydes: salicylaldehyde (H2L1), ovanillin (H2L2), pyridoxal (H2L3) and 2,6-diformyl-4-methylphenol (H3L4), and their corresponding Zn(II)complexes (1-4), are synthesized. All compounds are characterized by elemental analyses, infrared, UV-Vis, nuclear magnetic resonance spectroscopy and mass spectrometry. The structures of H2L2 and [Zn-2(L-1)(2)(H2O) (DMF)] (1a) (DMF = dimethylformamide) are solved by single crystal X-ray diffraction. The SB coordinates the metal center through the Ophenolate, Nimine and Sthiolate atoms. The radical scavenging activity is tested using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, with all ligand precursors showing IC50 values similar to 40 mu M. Cytotoxicity studies with several tumor cell lines (PC-3, MCF-7 and Caco-2) as well as a non-tumoral cell line (NHDF) are reported. Interestingly, 1 has relevant and selective antiproliferative effect against Caco-2 cells (IC50 = 9.1 mu M). Their antimicrobial activity is evaluated in five bacterial strains (Klebsiella pneumoniae, Acinetobacter baumannii, Listeria monocytogenes, Pseudomonas aeruginosa and Staphylococcus aureus) and two yeast strains (Candida albicans and Candida tropicalis) with some compounds showing bacteriostatic and fungicidal activity. The minimal inhibitory concentration (MIC90) of HnL against Mycobacterium tuberculosis is also reported, with H2L2 and H3L4 showing very high activity (MIC90 < 0.6 mu g/mL). The ability of the compounds to bind bovine serum albumin (BSA) and DNA is evaluated for H3L4 and [Zn-2(L-4)(CH3COO)] (4), both showing high binding constants to BSA (ca. 106 M 1) and ability to bind DNA. Overall, the reported compounds show relevant antitumor and antimicrobial properties, our data indicating they may be promising compounds in several fields of medicinal chemistry. A naphthalimide-pyridoxal conjugate 1 has been designed and synthesized. Compound 1 forms a stable greenish yellow colored gel in DMSO:H2O (8:1 v/v). Rheological study reveals that the gel is mechanically strong (G> G) over a wide range of applied strains. The morphology of the gel as determined by FESEM shows a highly cross-linked fibrous network. The gel is anion-responsive and is selectively transformed into a sol with a color change from greenish yellow to deep blue only in the presence of F- among other anions. In CH3CN, compound 1 was also sensitive to basic anions such as F- and AcO- ions. In solution, F- was differentiated from AcO- through a color change. While the yellow colored solution of 1 in acetonitrile was changed into deep blue in the presence of F-, AcO- ions gave a faint blue coloration. A similar colorimetric differentiation of F- from AcO- has been possible in CH3CN by a reusable Schiff base-linked Merrifield resin 1a or 1b.
