Koji amazake, prepared from rice koji, is a traditional Japanese sweet beverage. The main source of sweetness is glucose derived from rice starch following digestion by enzymes of Aspergillus oryzae during saccharification. The temperature of this process was empirically determined as 45 degrees C-60 degrees C, but no studies have systematically investigated the effect of temperature on saccharification efficiency. We addressed this in the present study by evaluating saccharification efficiency at various temperatures. We found that glucose content was the highest at 50 degrees C (100%) and was reduced at temperatures of 40 degrees C (66.4%), 60 degrees C (91.9%), and 70 degrees C (76.6%). We previously reported that 12 types of oligosaccharides are present in koji amazake; the levels of eight of these, namely nigerose, kojibiose, trehalose, isomaltose, gentiobiose, raffinose, panose, and isomaltotriose, were the highest at 50 degrees C-60 degrees C, whereas sophorose production was maximal at 70 degrees C. Based on these findings, we initially performed saccharification at 50 degrees C and then switched the temperature to 70 degrees C. The maximum amount of each saccharide including sophorose that was produced was close to the values obtained at these two temperatures. Thus, oligosaccharide composition of koji amazake is dependent on saccharification temperature. A naphthalimide-pyridoxal conjugate 1 has been designed and synthesized. Compound 1 forms a stable greenish yellow colored gel in DMSO:H2O (8:1 v/v). Rheological study reveals that the gel is mechanically strong (G> G) over a wide range of applied strains. The morphology of the gel as determined by FESEM shows a highly cross-linked fibrous network. The gel is anion-responsive and is selectively transformed into a sol with a color change from greenish yellow to deep blue only in the presence of F- among other anions. In CH3CN, compound 1 was also sensitive to basic anions such as F- and AcO- ions. In solution, F- was differentiated from AcO- through a color change. While the yellow colored solution of 1 in acetonitrile was changed into deep blue in the presence of F-, AcO- ions gave a faint blue coloration. A similar colorimetric differentiation of F- from AcO- has been possible in CH3CN by a reusable Schiff base-linked Merrifield resin 1a or 1b.

Aberrant activation of ERK signaling pathway usually leads to oncogenesis, and small molecular agents targeting this pathway are impeded by the emergence of drug resistance due to reactivation of ERK signaling. Compound DEL-22379 has been reported to inhibit ERK dimerization which was unaffected by drug-resistant mechanism reactivating the ERK signaling. Here, we discussed a structure-activity relationship study of DEL-22379. Forty-seven analogues were designed and synthesized. Each synthesized compound was biologically evaluated for their inhibitory rates on several tumor cell lines and compounds with high inhibitory rates were further evaluated for IC50 values. The structure-activity relationship of idolin-2-one scaffold and the impact of Z/E configuration on potency were discussed. The carbonylative Suzuki cross-coupling of 2-bromopyridine with various boronic acids to prepare unsymmetrical arylpyridine ketones has been carried out using palladium/N-heterocyclic carbene complexes as catalysts prepared in situ. The selectivity and the rate of these reactions are highly dependent on the conditions, i.e., nature of the palladium catalyst precursor, solvent, temperature and CO pressure. The main side-products arise from direct, non-carbonylative cross-coupling. Under the optimum conditions, arylpyridine ketones are recovered in high yields (60-88%). The antibacterial activities of the corresponding benzimidazole salts 2 were tested against Gram positive and negative bacteria using the agar dilution procedure, and their IC50 values have been determined.

The synthesis of a novel class of cyclometalated gold(III) complexes supported by benzoylpyridine, benzylpyridine, and (1R,2R)-(+)-1,2-diaminocyclohexane (DACH) ligands, along with their crystal structures, is reported. These compounds provide a new scaffold to investigate biological properties of gold(III) complexes. The six complexes were prepared and characterized, following reactions of (C,N) cyclometalated gold(III) scaffolds, [Au(C boolean AND N)Cl-2] with DACH, which yielded a new series of cyclometaled gold(III), 3-5, of the type [Au(C boolean AND NH)(DACH)(2)](+) and the nitrogen substituted cyclometalated Au(III), 6-8, of the type [Au-(C boolean AND N)(DACH)](2+). Antiproliferative activity of these complexes in a panel of cancer cells showed promising results with IC50 in the micromolar range and selectivity over normal epithelial cells, MRCS. Whereas 8 shows minimal interaction with superhelical DNA except at high gold concentrations of 500 mu M, complex 5 does not show interaction even at 1000 mu M. The complexes display significant uptake in OVCAR8 cancer cells within 200-1200 pmol/million cells with the exception of complex 4. Differential cellular uptake was observed for the complexes; for example, while 3 and 8 display significant uptake, 4 showed minimal uptake. The compounds proved to be stable under physiological conditions and were minimally affected by either glutathione or sodium ascorbate. Cell cycle studies reveal a G1 arrest induced by representative complexes. The results reveal that enhanced Au(III) stabilization promoted by combined cyclometalated and DACH ligands may offer ligand tuning insights for novel anticancer drug design.

A simple, and practical oxidative scission of aromatic olefins to carbonyl compounds using O-2 as the sole oxidant with poly(ethylene glycol) dimethyl ether as a benign solvent has been developed. A wide range of monosubstituted, gem-disubstituted, 1,2-disubstituted, trisubstituted and tetrasubstituted aromatic olefins was successfully converted into the corresponding aldehydes and ketones in excellent yields even with gram-scale reaction. Blue emitting phosphorescent materials with high efficiency and high stability are a key requirement for the wider adoption of organic light emitting devices (OLEDs). In order to achieve triplet derived emission at the higher energy region of the electromagnetic spectrum, a series of neutral, monocyclometalated gold(iii) complexes with trifluoromethyl and trifluoromethoxy functionalized 2-phenylpyridine (ppy) derivatives, 2-anilinopyridine (apy), 2-benzoylpyridine (bpy) and 2-benzylpyridine (bepy), as the cyclometalating framework and diaryl or monoaryl alkyne as ancillary ligands have been designed and synthesized. Extensive photophysical and chemical characterization by various nuclear magnetic resonance spectroscopy techniques, elemental analysis and single crystal X-ray diffraction studies of selected compounds was carried out to confirm the structural and the chemical identity of the complexes. The emission wavelength maxima of the complexes appear in the blue/sky blue region of the electromagnetic spectrum. Detailed photophysical investigations revealed that the different emission properties of the complexes are predominantly dictated by the electronic properties of the cyclometalating ligand. The interesting photoluminescence properties along with the facile synthetic access makes this a promising concept to obtain highly suitable blue emitting gold(iii) complexes.

Three novel ruthenium nitrosyl complexes [Ru(NO)Cl-3(InicMe)(2)] (1b), [RuNOCl3(NicEt)(2)] (1c) and [RuNOCl3(NicMe)(2)] (1d) (InicMe = methyl isonicotinate, NicEt = ethyl nicotinate, NicMe = methyl nicotinate)were prepared and crystal structure of the complexes were determined by single crystal XRD analysis. Finally, cytotoxicity of the compounds was tested on Hep2 and HepG2 cell lines. The combined influence of structural factors (Y, Z substituents) on the rate of catalyzed by Z-substituted pyridines reactions of phenyloxirane with Y-substituted N-aroylbenzenesulfonamides in acetonitrile at 293 K was studied. A cross-correlation analysis of the results of a multifactor kinetic experiment was carried out. A polylinear regression, that adequately describes the non-additive effects of substituents Y and Z, was calculated. Due to the interaction of the joint effects of structure, the cross-reaction series exhibits isoparametric properties. The mechanism of the catalytic process has been discussed.

New Delhi metallo-beta-lactamase-1 (NDM-1) is capable of hydrolyzing nearly all beta-lactam antibiotics, posing an emerging threat to public health. There are currently less effective treatment options for treating NDM-1 positive superbug, and no promising NDM-1 inhibitors were used in clinical practice. In this study, structure-activity relationship based on thiosemicarbazone derivatives was systematically characterized and their potential activities combined with meropenem (MEM) were evaluated. Compounds 19bg and 19bh exhibited excellent activity against 10 NDM-positive isolate clinical isolates in reversing MEM resistance. Further studies demonstrated compounds 19bg and 19bh were uncompetitive NDM-1 inhibitors with Ki = 0.63 and 0.44 mu mol/L, respectively. Molecular docking speculated that compounds 19bg and 19bh were most likely to bind in the allosteric pocket which would affect the catalytic effect of NDM-1 on the substrate meropenem. Toxicity evaluation experiment showed that no hemolysis activities even at concentrations of 1000 mg/mL against red blood cells. In vivo experimental results showed combination of MEM and compound 19bh was markedly effective in treating infections caused by NDM-1 positive strain and prolonging the survival time of sepsis mice. Our finding showed that compound 19bh might be a promising lead in developing new inhibitor to treat NDM-1 producing superbug.
