A series of novel 5-amide benzofuran derivatives I [ R = ethylsulfonyl, benzoyl, methoxysulfamoyl, cyclohexylcarbamoyl, etc.] were designed and synthesized as NLRP3-inflammasome inhibitors. Compound I [ R = 2-methylpropanoyl] showed slightly increased activity (IC50 = 3.85μM) against IL-1β release.
