Several palladium(ii) and platinum(ii) complexes (1-20) of general formula [M(L-n)(X)(Y)] [M = Pd, X = Y = Cl (1-Cl-4-Cl), X = Y = OAc (1-OAc-4-OAc); M = Pt: X = Y = Cl (5-8); M = Pd, X = Cl, Y = CH3 (9-12); M = Pt, X = Cl, Y = CH3 (13-16) or X = Y = CH3 (17-20); n = 1-4] have been synthesized by reaction of different Pd(ii) and Pt(ii) derivatives with various 3-substituted 1-(2-pyridyl)-imidazo[1,5-a]pyridines; i.e.L-n = 1-(2-pyridyl)-3-arylimidazo[1,5-a]pyridine (aryl = Phenyl, L-1; 2-o-Tolyl, L-2; Mesityl, L-3) and 1-(2-pyridyl)-3-benzylimidazo[1,5-a]pyridine (L-4). Detailed spectroscopic investigation (including IR, mono- and bi-dimensional H-1 NMR) and elemental analysis has been performed for all these species, allowing their complete characterization. L-n act as N,N-bidentate ligands and coordinate the metal centers in a chelate fashion through the pyridyl (N-py) and the pyridine-like nitrogen atom of the imidazo[1,5-a]pyridine group (N-im). The X-ray structural analysis performed on two of Pd(ii) and three Pt(ii) complexes, namely [Pd(L-2)(CH3)Cl] (10), [Pd(L-3)(CH3)Cl] (11) and [Pt(L-1)Cl-2] (5), [Pt(L-4)Cl-2] (8), [Pt(L-2)(CH3)Cl] (14) confirmed the spectroscopic and analytical data. Finally DFT studies unveiled the structural reasons behind the inertia of the synthesised compounds toward metalation, identified as the higher angle steric strain in comparison with the analogous bipyridine complexes.

A series of two new mono- and one binuclear m-nitrato bridged copper(II) complexes [Cu(L)(HL)]ClO4 (1), [Cu(HL)(NO3)(H2O)](2)NO3 center dot H2O (2) and [Cu-2(L-2)(mu-NO3)(2)] (3), with an unsymmetrical NNO donor Schiff base (HL) have been synthesized and characterized by elemental analysis, FTIR, CV, UV-vis and EPR spectroscopy. Their molecular structures were also determined by single crystal X-ray crystallography. In the binuclear complex 3, the Cu center dot center dot center dot Cu distance is 3.494 angstrom. In 1, 2 and 3, the Cu(II) centers have distorted square pyramidal geometry (tau(5) = 0.05-0.17). Evidence of weak pi center dot center dot center dot pi stacking intermolecular interactions along with other non-covalent interactions (hydrogen bonding) was observed by analyzing the respective crystal structures of the complexes. Thus, these hydrogen bonds, pi center dot center dot center dot pi stacking interactions and other weak intermolecular interactions establish in the form of supramolecular architectures a crystalline network environment. The non-covalent interactions were also investigated by employing Hirshfeld Analysis. The room temperature magnetic moments of the mononuclear complexes are less than the spin only values which are indicative of small interactions. Also, significant magnetic interactions were not exhibited by binuclear copper(II) complex 3 in the variable temperature magnetic measurements. The X-band EPR spectra of all three complexes exhibit copper(II) hyperfine structures as well as zero-field splitting which are appropriate for the triplet states of dimers. In complexes 1 and 2, the presence of pseudo dipolar interactions is proposed. Quantum chemical calculations (DFT) were carried out on complexes 1-3 to explore the electronic and spectral properties of these newly synthesized complexes. These complexes show significant antiproliferative and SOD activity. The SOD activity measured in terms of kMcCF is in the range 4.94-12.31 (mol L)(-1)s(-1))x 10(4). (C) 2021 Elsevier Ltd. The combined influence of structural factors (Y, Z substituents) on the rate of catalyzed by Z-substituted pyridines reactions of phenyloxirane with Y-substituted N-aroylbenzenesulfonamides in acetonitrile at 293 K was studied. A cross-correlation analysis of the results of a multifactor kinetic experiment was carried out. A polylinear regression, that adequately describes the non-additive effects of substituents Y and Z, was calculated. Due to the interaction of the joint effects of structure, the cross-reaction series exhibits isoparametric properties. The mechanism of the catalytic process has been discussed.

Steric hindrance in the binding pocket of an alcohol dehydrogenase (ADH) has a great impact on its activity and stereoselectivity simultaneously. Due to the subtle structural difference between two bulky phenyl substituents, the asymmetric synthesis of diaryl alcohols by bioreduction of diaryl ketones is often hindered by the low activity and stereoselectivity of ADHs. To engineer an ADH with practical properties and to investigate the molecular mechanism behind the asymmetric biocatalysis of diaryl ketones, we engineered an ADH from Lactobacillus kefiri (LkADH) to asymmetrically catalyse the reduction of 4-chlorodiphenylketones (CPPK), which are not catalysed by the wild type (WT) enzyme. Mutants seq1-seq5 with gradually increased activity and stereoselectivity were obtained through iterative shrinking mutagenesis. The final mutant seq5 (Y190P/I144V/L199V/E145C/M206F) demonstrated the highest activity and excellent stereoselectivity of >99% ee. Molecular simulation analyses revealed that mutations may enhance the activity by eliminating steric hindrance, inducing a more open binding loop and constructing more noncovalent interactions. The pro-R pose of CPPK with a halogen bond formed a pre-reaction conformation more easily than the pro-S pose, resulting in the high ee of (R)-CPPO in seq5. Moreover, different halogen bonds formed due to the different positions of chlorine substituents, resulting in opposite substrate binding orientation and stereoselectivity. Therefore, the stereoselectivity of seq5 was inverted toward ortho- rather than para-chlorine substituted ketones. These results indicate that the stereocontrol element of LkADH was changed to recognise diaryl ketones after steric hindrance was eliminated. This study provides novel insights into the role of steric hindrance and noncovalent bonds in the determination of the activity and stereoselectivity of enzymes, and presents an approach producing key intermediates of chiral drugs with practical potential.

Five new nickel(II) complexes [Ni(L)NCS] (1b), [Ni(L)N-3] (2), [Ni(L)N(CN)(2)] (3), [Ni(L)OAc] (4) and [Ni(HL)SO4] (5) of a thiosemicarbazone HL (where HL = 2-benzoylpyridine-N(4)-phenylthiosemicarbazone) including pseudohalides like thiocyanate, azide and dicyanamide have been synthesized and characterized by elemental analysis, magnetic susceptibility, FT-IR, electronic and NMR spectroscopic techniques. The interesting amido-iminol tautomerism enables the pro-ligand to exist in amido form as well as deprotonated iminol form thereby functioning as tridentate NNS donor, coordinating through pyridyl nitrogen atom, azomethine nitrogen atom and amido/iminolate sulfur atom in the complexes. The crystal structures were established by single crystal XRD analysis. Two different single crystals of the compound thiocyanato complex were obtained with and without DMF solvent molecule having triclinic and monoclinic lattices with P (1) over bar and P2(1)/n space groups respectively. Also, the compound 2 is having a monoclinic lattice with P21/n space group. Nickel(II) exists in distorted square planar geometry in all the complexes. Further DFT calculations were carried out at B3LYP/TZVP level of theory to obtain an insight on the stability and the nature of frontier orbitals. Finally, electrostatic potential plots mapped on optimized geometries substantiate the noncovalent interactions found in their respective crystal structures. (C) 2018 Elsevier Ltd. The carbonylative Suzuki cross-coupling of 2-bromopyridine with various boronic acids to prepare unsymmetrical arylpyridine ketones has been carried out using palladium/N-heterocyclic carbene complexes as catalysts prepared in situ. The selectivity and the rate of these reactions are highly dependent on the conditions, i.e., nature of the palladium catalyst precursor, solvent, temperature and CO pressure. The main side-products arise from direct, non-carbonylative cross-coupling. Under the optimum conditions, arylpyridine ketones are recovered in high yields (60-88%). The antibacterial activities of the corresponding benzimidazole salts 2 were tested against Gram positive and negative bacteria using the agar dilution procedure, and their IC50 values have been determined.

Herein, we report a graphene oxide (GO) catalyzed condensation of non-activated esters and amines, that can enable diverse amides to be synthesized from abundant ethyl esters forming only volatile alcohol as a by-product. GO accelerates ester to amide conversion in the absence of any additives, unlike other catalysts. A wide range of ester and amine substrates are screened to yield the respective amides in good to excellent yields. The improved catalytic activity can be ascribed to the oxygenated functionalities present on the graphene oxide surface which forms H-bonding with the reactants accelerating the reaction. Improved yields and a wide range of functional group tolerance are some of the important features of the developed protocol.

Synthesis of 2,6-disubstituted pyridines from pyridotriazoles through palladium-catalyzed aerobic oxidative denitrogenative reactions has been described. Denitrogenation of arylated pyridotriazoles generates metal-carbene intermediates in situ and provides selectively 6-aryl-2-benzoylpyridines. A new coumarin based fluorescent switch (HCBP) has been fabricated which displays high selective sensing towards Al3+ among other metal cations at physiological pH. On gradual addition of Al3+ specifically, HCBP shows a brilliant turn-on emission enhancement of about similar to 13-fold with about 50 nm red shift at 481 nm in MeOH/H2O (1/1, v/v) solution. A new cyan fluorescent probe, MIPY-DNBS, using an imidazo[1,5-alpha]pyridine derivative as the fluorophore and 2,4-dinitrobenzensufonate as the recognition site for the selective detection of thiols (Cys, GSH, and Hcy), was designed and synthesized.
