Pyridine-substituted alkylidenecyclopropanes (Py-ACPs) react with gold(III) salts under mild reaction conditions through an unprecedented, proximal ring-opening pathway, to generate highly appealing, catalytically active pyridine alkenyl [C<^>N]-gold(III) species. Mechanistic studies reveal that the activation of the C-C bond of the ACP takes place through an unusual concerted, sigma-bond metathesis type-process.

Flavin-dependent ‘ene’-reductases (EREDs) are highly selective catalysts for the asymmetric reduction of activated alkenes. This function is, however, limited to enones, enoates, and nitroalkenes using the native hydride transfer mechanism. Here we demonstrate that EREDs can reduce vinyl pyridines when irradiated with visible light in the presence of a photoredox catalyst. Experimental evidence suggests the reaction proceeds via a radical mechanism where the vinyl pyridine is reduced to the corresponding neutral benzylic radical in solution. DFT calculations reveal this radical to be dynamically stable, suggesting it is sufficiently long-lived to diffuse into the enzyme active site for stereoselective hydrogen atom transfer. This reduction mechanism is distinct from the native one, highlighting the opportunity to expand the synthetic capabilities of existing enzyme platforms by exploiting new mechanistic models.

Strained compounds are privileged moieties in modern synthesis. In this context, 1-azabicyclo[1.1.0]butanes are appealing structural motifs that can be employed as click reagents or precursors to azetidines. We herein report the first telescoped continuous flow protocol for the generation, lithiation, and electrophilic trapping of 1-azabicyclo[1.1.0]butanes. The flow method allows for exquisite control of the reaction parameters, and the process operates at higher temperatures and safer conditions with respect to batch mode. The efficiency of this intramolecular cyclization/C3-lithiation/electrophilic quenching flow sequence is documented with more than 20 examples.

The alkynes served as both alkylating and acylating agents via switching between anaerobic and aerobic conditions. A three-descriptor quantitative structure-activity/toxicity relationship (QSAR/QSTR) model was developed for the skin permeability of a sufficiently large data set consisting of 274 compounds, by applying support vector machine (SVM) together with genetic algorithm. The optimal SVM model possesses the coefficient of determination R-2 of 0.946 and root mean square (rms) error of 0.253 for the training set of 139 compounds; and a R-2 of 0.872 and rms of 0.302 for the test set of 135 compounds. Compared with other models reported in the literature, our SVM model shows better statistical performance in a model that deals with more samples in the test set. Therefore, applying a SVM algorithm to develop a nonlinear QSAR model for skin permeability was achieved.

A simple, and practical oxidative scission of aromatic olefins to carbonyl compounds using O-2 as the sole oxidant with poly(ethylene glycol) dimethyl ether as a benign solvent has been developed. A wide range of monosubstituted, gem-disubstituted, 1,2-disubstituted, trisubstituted and tetrasubstituted aromatic olefins was successfully converted into the corresponding aldehydes and ketones in excellent yields even with gram-scale reaction. Some control experiments were also conducted to support a possible reaction pathway.

Three new mononuclear complexes of the type [M(L)(2)](ClO4)(2), with Ni(II) (1), Cu(II) (2) and Zn(II) (3) ions, have been synthesized and structurally characterized, using the N,N,O donor Schiff base ligand 2-[(phenyl-pyridine-2-yl-methylene)-amino]-ethanol]. An X-ray diffraction study reveals identical crystallization patterns for each of the complexes, in the monoclinic crystal system with the C2/c space group. Interestingly, the synthesized complexes are isostructural in nature. The complexes showed biomimetic phenoxazinone synthase-like activity for the aerial oxidation of o-aminophenol to phenoxazine-2-one, separately in methanol (MeOH) and acetonitrile (ACN) solutions. For each of the complexes, the turnover numbers were found to be higher in MeOH than in ACN. The highest catalytic activity between among the three complexes was found to be for complex 2. Silver nanoparticles are generated from simple silver (I) salts and operate at 25 degrees C under 20 bar of hydrogen pressure. Various aliphatic and aromatic ketones, including natural products were reduced into the corresponding alcohols in high yields. This silver catalyst allows for the selective hydrogenation of ketones in the presence of other functional groups.

We report a mild palladium-catalyzed selective dehydrogenation of 4-alkylpyridines that exploits their soft enolization to alkylidene dihydropyridines using allyl chloroformate under metal-free conditions. Treatment of these intermediates with a palladium catalyst liberates an alkylpyridylic anion and an allylpalladium(II) intermediate, which combine and undergo beta-hydride elimination to install a double bond. Importantly, the formation of gas byproducts makes the process very practical. Furthermore, the reaction tolerates a broad range of functional groups, including many that can be dehydrogenated using similar palladium-catalyzed reactions, and is selective for 4-alkylpyridines even in substrates bearing multiple pyridylic positions.

Green and sustainable chemistry approaches necessitate an ongoing investigation towards new, environmentally benign and selective catalysts. We have thus prepared a new benzothiazole-scaffolded hydrazone Schiff base ligand L and coordinated it with copper(II) ions leading to five different complexes, the form of which in the solid state is counterion-dependent. Nitrate, chloride and trifiate anions lead to monometallic species, whereas utilization of sulfates and tetrafluoroborates is responsible for the formation of bimetallic assemblies. The catalytic efficiency of synthesized compounds was shown in Cu-TEMPO (2,2,6,6-tetramethyl-l-piperidinoxyl) aerobic oxidation of activated alcohols resulting in good to excellent conversions and 100% selectivity to form the corresponding aldehydes. Considering the experimental results and Density Functional Theory calculations we propose two most plausible reaction mechanisms that further corroborate the enhanced activity of bimetallic species. It appears that the tridentate character of the ligand sacrifices some of the overall catalytic efficiency for the selectivity of the process. Aqueous reaction medium, low catalyst loading, air as oxidant and exclusive oxidative selectivity render these Cu ll complexes promising candidates for further improvement. (C) 2020 Elsevier B.V.
